Rapid screening of staphylokinase protein variants using an unpurified cell‐free expression system

open access: yesFEBS Open Bio, EarlyView.
An unpurified cell‐free protein synthesis (CFPS) platform enables rapid functional screening of staphylokinase variants. Direct plasminogen‐activation assays performed in microplate format provide real‐time activity readouts, allowing rapid identification and ranking of variants with improved or reduced fibrinolytic activity without protein ...

Manifold Learning for Aerodynamic Shape Design Optimization

open access: yesAerospace
The significant computational cost incurred due to the iterative nature of Computational Fluid Dynamics (CFD) in traditional aerodynamic shape design frameworks poses a major challenge, especially in the context of modern integrated design requirements ...

A New Cokriging Method for Variable-Fidelity Surrogate Modeling of Aerodynamic Data [PDF]

open access: yes, 2010
Cokriging is a statistical interpolation method for the enhanced prediction of a less intensively sampled primary variable of interest with assistance of intensively sampled auxiliary variables. In the geostatistics community it is referred to as two- or

Large‐scale bidirectional arrayed genetic screens identify OXR1 and EMC4 as modifiers of αSynuclein aggregation

open access: yesFEBS Open Bio, EarlyView.
Activation of the mitochondrial protein OXR1 increases pSyn129 αSynuclein aggregation by lowering ATP levels and altering mitochondrial membrane potential, particularly in response to MSA‐derived fibrils. In contrast, ablation of the ER protein EMC4 enhances autophagic flux and lysosomal clearance, broadly reducing α‐synuclein aggregates.

YIPFα1A expression is regulated by multilayered molecular mechanisms

open access: yesFEBS Open Bio, EarlyView.
YIPFα1A, a five‐pass Golgi protein, is regulated at multiple layers. (1) Rare‐codon enrichment drives translation‐coupled mRNA decay. (2) A proximal 3′‐UTR element stabilizes mRNA. (3) A distal 3′‐UTR element included by alternate poly(A) site usage represses translation, which can be overridden by the proximal 3′‐UTR element.

An Improved Approach for Reducing the Dimensionality of Wing Aerodynamic Optimization Considering Longitudinal Stability

open access: yesAerospace
The wing aerodynamic shape optimization is a typical high-dimensional problem with numerous independent design variables. Researching methods to reduce the dimensionality of optimization from the perspective of aerodynamic characteristics is necessary ...

Numerical Airfoil Optimization Using a Reduced Number of Design Coordinates [PDF]

open access: yes
A method is presented for numerical airfoil optimization whereby a reduced number of design coordinates are used to define the airfoil shape. The approach is to define the airfoil as a linear combination of shapes.

Derivation and characterization of retinal pigment epithelium from urine‐derived iPSCs

open access: yesFEBS Open Bio, EarlyView.
Age‐related macular degeneration causes vision loss via RPE dysfunction and loss. Traditional iPSC therapies rely on invasive biopsies, limiting scalability. Here, we utilize urine‐derived stem cells as an accessible source to generate u‐iPSCs, successfully differentiated into pigmented RPE. This “Urine‐to‐Retina” platform provides a promising path for
