Mechanism‐informed machine learning for individualized tacrolimus dose adjustment in the early post‐kidney transplant period

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Abstract Aim Tacrolimus dosing in the early post‐kidney transplant period is challenging due to a narrow therapeutic index and substantial interindividual pharmacokinetic (PK) variability. This study aimed to develop and validate mechanism‐informed machine learning (ML) models to support individualized tacrolimus dosing during this critical period ...

Long‐term impact of Elexacaftor/Tezacaftor/ivacaftor on pulmonary, nutritional and metabolic outcomes in homozygous F508del cystic fibrosis patients: A real‐world cohort study

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aim Elexacaftor/tezacaftor/ivacaftor (ETI) has markedly improved cystic fibrosis (CF) outcomes. However, its long‐term impact on nutrition, metabolism and liver health remains underexplored. We assessed 30‐month changes in pulmonary, nutritional, metabolic and inflammatory markers in people with CF (PwCF) homozygous for F508del.

Effect of carbamazepine on the pharmacokinetics of vepdegestrant, a PROteolysis TArgeting Chimera estrogen receptor degrader, in healthy adults

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aim To evaluate the effects of carbamazepine, a strong cytochrome P450 (CYP)3A4 inducer, on the pharmacokinetics and safety of vepdegestrant, a PROteolysis TArgeting Chimera estrogen receptor degrader. Methods This was a phase 1, open‐label, fixed‐sequence, two‐period study in healthy adult participants.

Comparative efficacy of GLP‐1 RA, tirzepatide and SGLT‐2 inhibitors in metabolic liver disease: A network meta‐analysis

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aim Metabolic liver disease, including nonalcoholic fatty liver disease (NAFLD) and nonalcoholic steatohepatitis, is a major cause of chronic liver dysfunction worldwide, creating an urgent need for effective treatments. This systematic literature review (SLR) and network meta‐analysis (NMA) systematically reviews and compares the efficacy and safety ...

Population pharmacokinetic/pharmacodynamic modelling to evaluate favipiravir in combination with lopinavir–ritonavir in patients with COVID‐19

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aims The repurposed use of favipiravir in COVID‐19 has been reported to have limited clinical efficacy, yet it has been widely used in some countries. Favipiravir causes mutagenesis in RNA viruses, and it is currently unknown whether it may have a measurable effect on the virus in humans.

Inhibition of Alanine Transaminase by the Hypoglycæmic Sulphonylurea Derivatives

Nature, 1957
BECAUSE of the recent interest in the mechanism of action of the two antidiabetic sulphonylurea derivatives, carbutamide (N1 sulphanilyl N2 n-butylcarbamide) and tolbutamide (N1 p-toluylsulphonyl N2 n-butylcarbamide), the effect of these compounds on the liver transaminase system was investigated, since inhibition of transamination reactions could lead

Aspartate transaminase/alanine transaminase (De Ritis ratio) predicts survival in major burn patients

Burns, 2022
Although treatment of burn patients has significantly improved in recent decades, major burns remain fatal. Therefore, the evaluation of the death risk of the patients with extensive burns is very important. The ratio between the serum levels of aspartate transaminase and alanine transaminase (De Ritis ratio) was an independent predictor of poor ...

Aspartate and Alanine Transaminases in Middle Ear Effusions

Acta Oto-Laryngologica, 1975
Mucous secretion from 24 ears with secretory otitis media was analysed for aspartate aminotransferase (Asp-AT) and from 18 ears for alanine aminotransferase (Ala-AT). The activity of both these enzymes in middle ear fluids was 40 times larger than in serum, the differences being statistically significant.

Comparative properties of glutamic-alanine transaminase from several sources

Archives of Biochemistry and Biophysics, 1964
Abstract Rat liver mitochondrial glutamic-alanine transaminase can be distinguished from the supernatant enzyme on the basis of its markedly greater lability and its nonresponsiveness to glucocorticoid administration. The activity of this enzyme in rat skeletal muscle, heart, and kidney are considerably less than that in liver and are also ...
