Direct Pharmacological Activation of AMPK Extends Lifespan in Yeast, Worms and Flies

open access: yesAging Cell, Volume 25, Issue 10, October 2026.
AMP‐activated protein kinase (AMPK) is a key evolutionarily conserved sensor of energy homeostasis, which is implicated in metabolic health and disease, as well as in ageing. Here, we show that direct pharmacological activation of AMPK with the compound 991 in vivo extends lifespan in multiple model organisms: the fission yeast Schizosaccharomyces ...

Meta‐Analysis: Enhanced Liver Fibrosis (ELF) Test for Identifying Significant Fibrosis, Advanced Fibrosis, and Cirrhosis in Patients With Metabolic Dysfunction‐Associated Steatotic Liver Disease

open access: yesAlimentary Pharmacology &Therapeutics, Volume 64, Issue 7, Page 864-889, October 2026.
A metaanalysis of 34 studies/37 cohorts using the Siemens Healthineers ELF test for biopsy‐staged F2, F3, or F4 MASLD‐associated fibrosis validates 9.0 (≥F2), 9.8 (≥F3), and 11.3 (F4) cut‐offs, and establishes 10.5 as a high‐specificity F4 cut‐off. This potentially holds implications for guideline revisions and aiding qualification for antifibrotic ...

Transcriptomic and metabolomic analyses reveal that lobeglitazone ameliorates hepatic steatosis in rats concurrent with regulation of mitochondrial pyruvate metabolism

open access: yesBritish Journal of Pharmacology, Volume 183, Issue 19, Page 5935-5952, October 2026.
Background and Purpose Lobeglitazone is a thiazolidinedione and PPARγ agonist that improves metabolic parameters and hepatic steatosis, but its mechanisms are not fully understood. This study investigated the effects of lobeglitazone on hepatic transcriptomic and metabolic profiles in a rat model of obesity and Type 2 diabetes mellitus.

Discovery of Epidermal Growth Factor‐Like 7 as a Potential Non‐Invasive Biomarker for Fibrosis and Mortality Risk in MASLD

open access: yesClinical and Translational Science, Volume 19, Issue 10, October 2026.
Proteomic discovery and validation of EGFL7 as a potential non‐invasive biomarker in MASLD. EGFL7 was identified as the top‐ranked proteomic feature in a clinical cohort, demonstrated diagnostic performance comparable to FIB‐4 for fibrosis assessment, and was associated with increased all‐cause mortality risk in the UK Biobank.
