Cyclodextrin Aldehydes are Oxidase Mimics

ChemBioChem, 2009
AbstractCyclodextrins containing 6‐aldehyde groups were found to catalyse oxidation of aminophenols in the presence of hydrogen peroxide. The catalysis followed Michaelis–Menten kinetics and is related to the catalysis previously observed with cyclodextrin ketones.

Identification of aldehyde oxidase 1 and aldehyde oxidase homologue 1 as dioxin-inducible genes

Toxicology, 2005
Aldehyde oxidases are a family of highly related molybdo-flavoenzymes acting upon a variety of compounds of industrial and medical importance. We have identified aldehyde oxidase 1 (AOX1) as a 2,3,7,8-tetrachlorodibenzo-p-dioxin (dioxin) inducible gene in the mouse hepatoma cell line Hepa-1.

Metabolism of Xenobiotics by Aldehyde Oxidase

Current Protocols in Toxicology, 2015
AbstractAldehyde oxidase (AO) is a cytosolic molybdoflavoprotein whose contribution to the metabolism and clearance of xenobiotics‐containing heterocyclic rings has attracted increased interest in recent years. This unit details methods for identification and confirmation of AO as a metabolic pathway as well as a method for estimating clearance of ...

A comparison of the specificities of xanthine oxidase and aldehyde oxidase

Archives of Biochemistry and Biophysics, 1972
Abstract This study directly compares the specificities of the structurally similar hydroxylating enzymes, aldehyde oxidase and xanthine oxidase. Michaelis-Menten constants for a variety of substrates of xanthine oxidase were in general lower than those of aldehyde oxidase. With respect to the rates of oxidation, the basic similarity was a preference

Liver aldehyde oxidase: A form of xanthine oxidase

Biochimica et Biophysica Acta, 1960
Abstract Pig liver aldehyde oxidase is not specific for aldehydes, but in addition catalyzes the oxidation at reduced rates of hypoxanthine, xanthine and DPNH. The sites of binding of acetaldehyde and hypoxanthine by the enzyme appear to be different.

Liver aldehyde oxidase and xanthine oxidase genetics in the mouse

Animal Blood Groups and Biochemical Genetics, 1981
SummaryA ‘null’ activity variant for the major liver isozyme of aldehyde oxidase (AOX‐1) in adult male mice and an electrophoretically distinct, high activity variant of the second liver isozyme (AOX‐2) were used to examine the segregation of the genetic loci encoding these enzymes (Aox‐1 and Aox‐2 respectively) in breeding studies.

The purification and properties of aldehyde oxidase

Biochimica et Biophysica Acta, 1962
Abstract 1. 1. Highly purified aldehyde oxidase has been prepared from pig liver by reverse ammonium sulphate fractionation, heat treatment and chromatography on DEAE- cellulose and hydroxyl-apatite. 2. 2. Although pure by electrophoresis the enzyme shows several minor components in the ultracentrifuge. The flavin content is 5 mμmoles/mg, and

Structure and function of mammalian aldehyde oxidases

Archives of Toxicology, 2016
Mammalian aldehyde oxidases (AOXs; EC1.2.3.1) are a group of conserved proteins belonging to the family of molybdo-flavoenzymes along with the structurally related xanthine dehydrogenase enzyme. AOXs are characterized by broad substrate specificity, oxidizing not only aromatic and aliphatic aldehydes into the corresponding carboxylic acids, but also ...

Cyclophosphamide potentiation and aldehyde oxidase inhibition by phosphorylated aldehydes and acetals

Journal of Medicinal Chemistry, 1980
Fourteen phosphorylated acetals and aldehydes were synthesized for testing in vitro as inhibitors or substrates of aldehyde oxidase, an enzyme involved in the conversion of aldophosphamide to inactive carboxyphosphamide, and for concurrent in vivo administration with cyclophosphamide to mice bearing L1210 ascites tumor cells.

Inhibition of xanthine oxidase by various aldehydes

Biochemistry, 1984
The inactivation of bovine milk xanthine oxidase by various aldehydes has been investigated. For each aldehyde, the inactivation reaction gives rise to a unique molybdenum(V) electron paramagnetic resonance signal from xanthine oxidase (the Inhibited signal). Of the aldehydes tested, only a few (mainly aromatic) failed to undergo this reaction.
