Aminopterin (4-a m i n o-pteroylglutamic acid) is a potentially dangerous drug. It can favorably affect the lesions of psoriasis regularly, but the range between beneficial and toxic doses is narrow. The possibility of irreversible bone marrow aplasia is of chief concern in toxicity.

Antifolics are rational for psoriasis. Normal renal clearance is of vital importance; this may be true of normal liver functions as well. No fatal or near fatal reactions occurred in our series. We have reviewed two deaths following aminopterin (Aminopterin Sodium) therapy, apparently from inadequate renal clearance, one severe reaction for the same ...

Maternal Aminopterin Ingestion
Archives of Pediatrics & Adolescent Medicine, 1973To the Editor. —We read with interest the case report by E. B. Shaw, MD: "Fetal Damage Due to Maternal Aminopterin Ingestion: Follow-Up at Age 9 Years," as well as the editorial comment by Dr. Warkany (Am J Dis Child124:93-94, 1972). We suspect that the patient's apparently good current mental development and the mildness of the earlier mental defect ...

Teratogenic effects of aminopterin in sheep
Teratology, 1968AbstractSheep were given subcutaneous injections of aminopterin at different times and for different intervals during gestation. Congenital malformations occurred after treatment at all periods. The principal malformations observed were rotational and flexing deformities of the appendicular skeleton, kyphosis, scoliosis, torticollis, aplasia of the ...

Caenorhabditis briggsae: Effects of aminopterin
Experimental Parasitology, 1974Abstract The characteristic features of senescence developed prematurely in Caenorhabditis briggsae treated with the DNA synthesis inhibitor aminopterin at the minimum dosage which inhibits gonad formation. In addition, aminopterin induced other changes which thus far have not been associated with normal aging of C. briggsae .

The relative toxicities of methotrexate and aminopterin
Biochemical Pharmacology, 1966Abstract Hepatic aldehyde oxidases from rabbit, mouse, rat, and guinea pig have been purified 10-fold to 15-fold and shown to catalyze the oxidation of methotrexate to 7-hydroxymethotrexate. The mouse and rat liver enzymes, however, were unable to catalyze the oxidation of the related compound aminopterin, and the aminopterinoxidizing ability of the ...

JAMA, 1962
To the Editor:— It was called to our attention ( JAMA 178 :46 [Oct. 21] 1961) that a misstatement had been made in a correction. In this notation Dr. Kushner commented on an article, Treatment of Reticulosis ( JAMA 175 :84 [Jan. 14] 1961), wherein the dosage of aminopterin was incorrectly stated. We concur that this dosage is incorrect. However, Dr.

Methotrexate vs. Aminopterin for Psoriasis
Archives of Dermatology, 1961The value of antimetabolites for psoriasis was first observed by Gubner. 1 He pointed out that the suppressive effects of aminopterin were much greater on epithelium than on blood-forming organs. Aminopterin (4-aminopteroylglutamic acid) is a metabolically useless analogue of folic acid.

The effect of aminopterin on diphosphopyridine nucleotide
Archives of Biochemistry and Biophysics, 1954Abstract 1. 1. The administration of small doses of aminopterin to adult female rats over a period of several days, or a single large dose, resulted in a marked decrease in liver diphosphopyridine nucleotide (DPN) levels when the animals were sacrificed within a few hours following injection. 2. 2.
