The mechanism of myeloperoxidase-catalysed oxidation of aminopyrine

1. Myeloperoxidase catalysed the H2O2-supported oxidation of aminopyrine in the presence of Cl-, generating the aminopyrine cation radical (AP+.). The rate of AP+. formation was determined by monitoring the absorbance at 565 nm. The pH optimum of the reaction was around 5.0. The rate of AP+.

Kinetics of the nitrosation of aminopyrine to give dimethylnitrosamine

The initial rate of nitrosation of aminopyrine (AP, Pyramidon) to give dimethyl nitrosamine (DMN) showed maxima at pH 2.0 and 3.1. At pH 2, initial rate was proportional to nitrite concentration squared down to 6 mM, below which it decreased rapidly.

Abstract A study has been made of age and maturational determinants as well as of the incubation conditions which effect aminopyrine demethylase activity of rat liver. The specific activity of N -demethylation in vitro by the 9000 g supernatant fraction increased about 3-fold in the first 30 days after birth.
