This study provides insight into the dynamic equilibrium mechanism of a novel structural variant, small inverted triplication (SIT), which is generated by misalignment of the 3’ flap generated under DNA replication stress with palindromic sequence. Engineering a Virus‐Derived X Family DNA Polymerase FvPolX for de novo DNA Synthesis

In this study, a Faustovirus‐derived polymerase (FvPolX) is engineered based on structural and sequence analysis of PolX enzymes. The optimal mutants demonstrate substantially enhanced catalytic activity toward dNTPs and 3′‐ONH2‐dNTPs compared to the wild‐type enzyme.
