Processing of Amyloid Precursor Protein and Amyloid Peptide Neurotoxicity
Current Alzheimer Research, 2008Alzheimer's disease is characterized by the presence of two types of lesions in brain: neurofibrillary tangles and senile plaques. Intraneuronal neurofibrillary tangles are made of paired helical filaments containing hyperphosphorylated microtubule associated protein tau.

Recent studies indicate that the deposition of beta-amyloid (Abeta) is related in the pathogenesis of Alzheimer's disease (AD), but the underlying mechanism is still not clear. The abnormal interactions of Abeta with metal ions such as copper are implicated in the process of Abeta deposition and oxidative stress in AD brains.

β-Amyloid precursor is a PEST protein
Biochemical and Biophysical Research Communications, 1989The beta-amyloid peptide is generated by proteolytic processing of a family of beta-amyloid precursor proteins. Here we report that beta-amyloid precursor proteins have a primary structure motif known as a PEST sequence, which is predictive of the class of most protease-sensitive rapidly turning over proteins.

Stimulated platelets release amyloid β-protein precursor
Biochemical and Biophysical Research Communications, 1990Human platelets can be stimulated by thrombin or ionomycin to secrete soluble truncated amyloid beta-protein precursor and particulate membrane fragments which contain C-terminal and N-terminal immunoreactive amyloid beta-protein precursor. This suggests a possible circulating source of beta-protein in serum which may play a role in the formation of ...
