Anaplasia Is Rare and Does Not Influence Prognosis in Adult Medulloblastoma [PDF]

open access: yesJournal of Neuropathology and Experimental Neurology, 2005
Histopathologic grading based on increasing anaplasia predicts clinical behavior of pediatric medulloblastomas. The present study was aimed at grading 86 medulloblastomas of adult patients (aged 18 and older) by anaplasia and analyzing the predictive power.

In vitro and in silico modelling of ROS1‐positive non‐small cell lung cancer reveals fusion‐dependent tyrosine kinase inhibitor responses

open access: yesMolecular Oncology, EarlyView.
Drug resistance limits treatment success in a subset of lung cancers driven by ROS1 gene alterations. Using patient‐derived cells and computer simulations, we studied three key mutations and how they affect five targeted drugs. The mutations reduced drug effectiveness in different ways by altering protein structure and behavior.

Risk Prediction Models for Recurrence After Curative Treatment of Early‐Stage or Locally Advanced Lung Cancer: A Systematic Review

open access: yesAging and Cancer, EarlyView.
This systematic review synthesizes prognostic models for survival and recurrence in resected non‐small cell lung cancer. While many models demonstrate moderate to good discrimination, few are externally validated and reporting quality is variable, limiting clinical applicability and highlighting the need for robust, transparent model development ...

LINC01605 Suppresses Esophageal Squamous Carcinogenesis by Regulating Squamous Cell Differentiation, Proliferation, and Migration

open access: yesFrontiers in Bioscience-Landmark
Background: Esophageal squamous cell carcinoma (ESCC) develops through a multistage process in which normal epithelium transitions into intraepithelial neoplasia and ultimately into invasive carcinoma. Elucidating the molecular changes

Surfaceome Reprogramming of Stemsomes Promotes Lung Cancer Targeting via Potentiated Receptor–Ligand Interactions

open access: yesAdvanced Science, EarlyView.
This study presents a surfaceome‐reprogramming strategy for mutation‐independent lung cancer therapy by repurposing dexamethasone to prime mesenchymal stem cell‐derived nanovesicles. The engineered vesicles leverage multi‐valent interactions mediated by upregulated adhesion proteins, EPHA2, and NOTCH3.

ANXA2+ Small Extracellular Vesicles Drive Chemoresistance in Anaplastic Thyroid Cancer by Promoting XRCC5 Lactylation and Enhancing Non‐Homologous End‐Joining Repair

open access: yesAdvanced Science, EarlyView.
ANXA2+ sEVs promote cisplatin resistance in ATC by stabilizing the SRC/LDHA interaction, increasing LDHA phosphorylation and activity, and lactate production. Elevated lactate levels promote KAT5‐mediated lactylation of XRCC5 at lysine 265, enhancing its binding to XRCC6.

Clinical Risk Factor for Detected Distant Metastasis and Anaplastic Transformation After Reoperation in Recurrent/Persistent Differentiated Thyroid Cancer: A Retrospective Cohort Study

open access: yesLaryngoscope Investigative Otolaryngology
Objectives Research on detected distant metastasis or anaplastic transformation in recurrent/persistent differentiated thyroid cancer is limited. We analyzed the clinical factors associated with detected distant metastasis or anaplastic transformation ...

Histone H3K18 Lactylation Promotes the Malignant Progression of Wilms Tumor via a PSRC1/AKT/HIF‐1α Positive Feedback Loop

open access: yesAdvanced Science, EarlyView.
In nephroblastoma, aberrant glycolysis drives lactate accumulation, which elevates histone H3K18 lactylation via p300. Lactylation of the PSRC1 promoter activates its transcription. PSRC1 competitively binds AKT, relieving PTEN‐mediated inhibition and triggering AKT/mTOR/HIF‐1α signaling.

Systemic Nanomechanical Single‐Cell Profiling Reveals Mechanophenotype Transitions Under Therapeutic Perturbation

open access: yesAdvanced Science, EarlyView.
Single‐cell mechanomics demonstrates that pharmacological perturbation changes cytoskeletal and cortical structure, suppressing cancer cell invasiveness. By integrating atomic force microscopy (AFM)‐based cortical measurements with stimulated emission depletion (STED)‐resolved adhesion and cytoskeletal organization, this approach identifies nanoscale ...
