Engineering Highly Cellularized Living Materials via Mechanical Agitation

A mechanical agitation strategy is developed to engineer highly cellularized living materials, achieving cell densities of up to 1 billion cells per milliliter. By precisely tuning properties such as stiffness and toughness in blood clots, the approach is validated in both in vitro and in vivo studies.

A dual‐head airbrush‐based spraying device, combined with a fibrinogen bioink enriched with glycosaminoglycans and collagen, enables the delivery of biomaterials and cells to promote wound healing. This system demonstrates effective skin regeneration in vitro and in vivo, with comparable results to autografts.

Rolling ultraminiaturized microneedle spheres (RUMS), created via two‐photon polymerization, unlock effective and painless topical delivery of messenger RNA (mRNA) and drugs through the skin. By gently forming thousands of microscopic pores, RUMS boost skin permeability by up to 100‐fold.
