Dimethyl fumarate combined with cisplatin at subcytotoxic doses sensitizes cervical cancer toward ferroptosis and apoptosis through GSH restriction and p53 (re)activation

Dimethyl fumarate (DMF) reduces growth of HPV‐positive cervical cancer spheroids and induces ferroptosis in cervical cancer cells via blocking SLC7A11/Glutathione (GSH) axis. Combination of subcytotoxic doses of DMF and cisplatin (CDDP) further suppresses spheroid growth and drives cell death in 2D culture models.

Efficacy and Mechanism of Action of Saireito Therapy for Autoimmune Recurrent Abortion Indexed by Antinuclear Antibody and Anticardioripine Antibody

妊娠12週以内の初期流産を3回以上繰り返した,ANA,抗カルジオリピン抗体(ACA IgG, IgM)陽性習慣流産87例に柴苓湯療法を行い,抗体価(量)の推移を検討した。この結果,妊娠例(49例)の流産阻止率は63.3%,ANA陽性例(32例)の流産阻止率は65.6%,ACA IgG, IgM(fetal calf serum使用ELISA法)陽性(29例)は65.5%,両抗体陽性例(12例)は75.0%であった。ANAに対しては有意な低下作用は認められなかったが,ACA IgMに対しては有意な低下作用が認められた。文献的に柴苓湯の有効作用はTh1/Th2サイトカインバランス調整作用による液性免疫の抑制作用によると推察された。しかし,ACAが低下しなかった二生児獲得例が存在する事実から ...

COMP–PMEPA1 axis promotes epithelial‐to‐mesenchymal transition in breast cancer cells

This study reveals that cartilage oligomeric matrix protein (COMP) promotes epithelial‐to‐mesenchymal transition (EMT) in breast cancer. We identify PMEPA1 (protein TMEPAI) as a novel COMP‐binding partner that mediates EMT via binding to the TSP domains of COMP, establishing the COMP–PMEPA1 axis as a key EMT driver in breast cancer.

IMPDH inhibition enhances cytarabine efficacy in SAMHD1‐expressing leukaemia cells via guanine nucleotide depletion

Cytarabine is a key therapy for acute myeloid leukaemia (AML), but its efficacy is limited by the dNTPase SAMHD1, which hydrolyses its active metabolite. Screening nucleotide biosynthesis inhibitors revealed that IMPDH inhibitors selectively sensitise SAMHD1‐proficient AML cells to cytarabine.
