Arachidonate 12-lipoxygenases with reference to their selective inhibitors

Lipoxygenase is a dioxygenase recognizing a 1-cis,4-cis-pentadiene of polyunsaturated fatty acids. The enzyme oxygenates various carbon atoms of arachidonic acid as a substrate and produces 5-, 8-, 12- or 15-hydroperoxyeicosatetraenoic acid with a conjugated diene chromophore.

Gossypol, a potent inhibitor of arachidonate 5- and 12-lipoxygenases

Gossypol inhibited 5- and 12-lipoxygenases of rat basophilic leukemia (RBL-1) cells with ID50 of 0.3 microM and 0.7 microM, respectively. Nearly two orders of magnitude of higher concentration of gossypol was required to inhibit prostaglandin synthetase. The inhibition was of a non-competitive type with respect to arachidonate.

Thromboxane A2 is well known as a proaggregatory and vasoconstrictive compound [1]. It is produced from arachidonic acid in platelets, and fatty acid cyclooxygenase responsible for the thromboxane synthesis is localized in the microsomal fraction of human platelets [2]. There is another metabolic pathway of arachidonic acid in human platelets.

Incubation of rat lung cytosol with arachidonic acid produced 12-hydroxy-5,8,10,14-eicosatetraenoic acid as a major product, which was identified by gas chromatography-mass spectrometry. By ammonium sulfate fractionation and DEAE-cellulose chromatography the arachidonate 12-lipoxygenase was purified about 30-fold from the rat lung cytosol.

Arachidonate 12-lipoxygenase of porcine leukocytes, which was purified to homogeneity by immunoaffinity chromatography, was analyzed for iron content by atomic absorption spectrophotometry. The enzyme contained 0.70 +/- 0.09 g atom of iron per mol of enzyme (mean +/- S.D., n = 4).
