Integrating Three Views of Arf1 Activation Dynamics
Journal of Molecular Biology, 2004The proteins Arno and Gea2 of the Sec7 family can promote GDP-GTP exchange on Arf1, a small GTP-binding protein, which coordinates coated vesicle formation for protein transport within the cell. Crystal structures of the essential Sec7 domain (Sec7d) of Gea2 in the free and Arf1-bound forms suggest that conformational dynamics of the Sec7d as well as ...

Preparation of Myristoylated Arf1 and Arf6
2005Arf proteins are members of the Arf family of small Ras-like GTP binding proteins. Six Arfs, grouped into three classes, have been identified in mammalian cells and three members have been identified in yeasts. Arf1 and Arf6, more extensively studied than other Arfs, have been found to affect membrane traffic and actin remodeling.

1995
Abstract Bovine ARF1 was cloned by screening a bovine adrenal chromaffin cDNA library with an oligonucleotide probe, derived from partial protein sequencing of purified bovine brain ARF (Sewell and Kahn 1988; accession number J03275).

ARF1 is directly involved in dynamin-independent endocytosis
Nature Cell Biology, 2007Endocytosis of glycosylphosphatidyl inositol (GPI)-anchored proteins (GPI-APs) and the fluid phase takes place primarily through a dynamin- and clathrin-independent, Cdc42-regulated pinocytic mechanism. This mechanism is mediated by primary carriers called clathrin-independent carriers (CLICs), which fuse to form tubular early endocytic compartments ...

In Vitro Assays of Arf1 Interaction with GGA Proteins
2005ADP-ribosylation factor 1 (Arf1) is a GTP-binding protein that regulates membrane traffic. This function of Arf1 is, at least in part, mediated by Arf1 x GTP binding to coat proteins such as coatomer, clathrin adaptor protein (AP) complexes 1 and 3, and gamma-adaptin homology-Golgi associated Arf-binding (GGA) proteins.

An Arf1 GTPase mutant with different responses to GEF inhibitors
Biochemical and Biophysical Research Communications, 2008Guanine nucleotide exchange factors (GEFs) stimulate the activation of small GTP-binding proteins (GTPases). Establishing their specificity is a challenging issue, in which chemical genetics are rapidly gaining interest. We report a mutation in the Arf1 GTPase, K38A, which differentially alters its sensitivity to GEF inhibitors.

ARF1 regulatory factors and COPI vesicle formation
Current Opinion in Cell Biology, 2002Understanding the biosynthetic pathways involved in cellular transport is an important issue in cell biology. More than a hundred years after the discovery of the Golgi apparatus, we still do not understand the regulation of vesicular transport to, within and from the Golgi apparatus.

ARF1, a Transcription Factor That Binds to Auxin Response Elements
Science, 1997The plant hormone auxin regulates plant physiology by modulating the interaction of transcription factors with auxin response elements (AuxREs) of the affected genes. A transcription factor, Auxin Response Factor 1 (ARF1), that binds to the sequence TGTCTC in AuxREs was cloned from Arabidopsis by using a yeast one ...
