Unpacking ‘Artemisinin Resistance’

Trends in Pharmacological Sciences, 2017
Artemisinin and its derivatives, in combination with partner drugs, are currently the most effective treatments for malaria parasite infection. Even though artemisinin has been widely used for decades, its mechanism of action had remained controversial until recently.

Artemisinin: mechanisms of action, resistance and toxicity

International Journal for Parasitology, 2002
Artemisinin and its derivatives are widely used throughout the world. The mechanism of action of these compounds appears to involve the heme-mediated decomposition of the endoperoxide bridge to produce carbon-centred free radicals. The involvement of heme explains why the drugs are selectively toxic to malaria parasites.

Pharmacotherapy for artemisinin-resistant malaria

Expert Opinion on Pharmacotherapy, 2021
Malaria, the most devastating parasitic disease, is currently treated with artemisinin-based combination therapies (ACTs). Unfortunately, some ACTs are unable to rapidly clear Plasmodium falciparum parasites from the blood stream and are failing to cure malaria patients; a problem, so far, largely confined to Southeast Asia.

The many paths to artemisinin resistance in Plasmodium falciparum.

Trends in Parasitology, 2023
Emerging resistance against artemisinin (ART) poses a major challenge in controlling malaria. Parasites with mutations in PfKelch13, the major marker for ART resistance, are known to reduce hemoglobin endocytosis, induce unfolded protein response (UPR ...

A Kelch13-defined endocytosis pathway mediates artemisinin resistance in malaria parasites

Science, 2020

An Update on Artemisinin Resistance

2019
While the precise mode of action of artemisinin (ART) derivatives remains obscure, it is nonetheless commonly accepted that ART generates reactive oxygen intermediates that contribute to cell death. Also, numerous studies confirm that point mutations in the propeller domain of K13 protein play a key role in resistance to ART derivatives. Because of its

Spread of Artemisinin Resistance in Malaria

New England Journal of Medicine, 2014
To the Editor: Ashley et al. (July 31 issue)1 recently described the spread of Plasmodium falciparum resistant to artemisinin across Southeast Asia but not Africa. This and other studies2,3 included patients with uncomplicated malaria, whereas young children with severe malaria in Africa represent a key population of interest with the highest mortality
