Inhibition of glutamate-aspartate transaminase by β-methylene-DL-aspartate

beta-Methylene-DL-aspartate, a new beta, gamma-unsaturated amino acid, is an irreversible inhibitor of soluble pig heart glutamate-aspartate transaminase (Ki approximately 3 mM with respect to the L-form; limiting rate constant for inactivation approximately 0.4 min-1).

Purification of glutamic-aspartic transaminase

Abstract A scheme for the preparation of a highly purified glutamic-aspartic transaminase is reported. The methods employed are, precipitation with acetone, ion-exchange chromatography on carboxymethyl cellulose, and zone electrophoresis. A purification degree of 50 is achieved and the enzyme obtained is the holoenzyme.

Tryptic hydrolysates of denaturated glutamic‐aspartic transminase fractionated on a Sephadex G‐25 column, gave an elution diagram containing six main fractions. When peptide maps were developed from the transaminase digest 37 spots were detected on the paper sheet.

Aspartate and Alanine Transaminases in Middle Ear Effusions

Mucous secretion from 24 ears with secretory otitis media was analysed for aspartate aminotransferase (Asp-AT) and from 18 ears for alanine aminotransferase (Ala-AT). The activity of both these enzymes in middle ear fluids was 40 times larger than in serum, the differences being statistically significant.
