Ciclopirox Olamine Inhibits the NLRP3 Inflammasome to Alleviate Inflammatory Diseases

open access: yesAdvanced Science, EarlyView.
There is no drug targeting the NLRP3 inflammasome that has been approved for use in clinical settings. Ciclopirox olamine (CPX), an antifungal agent approved by the US Food and Drug Administration (FDA), is identified as a specific and potent NLRP3 inflammasome inhibitor. CPX targets the NACHT domain of NLRP3 at Y381 to impair NLRP3 oligomerization and

Senescent Cell Derived Artificial Vesicle‐Based Senolytic Sonovaccine Platform with Augmented Lymph Node Delivery and Antigen Cross‐Presentation Efficacy

open access: yesAdvanced Science, EarlyView.
A senolytic sonovaccine platform (SenoVac) is developed, in which senescent cell‐derived vesicles serve as broad senescent cell antigen reservior. The “2‐step” click chemistry strategy for effective lymph node delivery, and ultrasound‐triggered endosomal escape to boost cross‐presentation, ensures efficient senescent cell clearance and disease ...

Cabrol Procedure in Complex Aortic Root Reconstruction: A Case Series of Three Young Patients With Acute Aortic Syndrome

open access: yesClinical Case Reports
Acute aortic dissection is a rare but life‐threatening syndrome, being accompanied by a mortality rate of 1%–2% per hour after the onset of symptoms if they remain untreated.

ApoE gene therapy: an overview and update

open access: yes, 2005
Atherosclerosis remains the leading cause of death in industrialized societies. Apolipoprotein E (ApoE) is an attractive candidate to treat hypercholesterolemia and coronary heart disease, as it is a circulating protein with pleiotropic ...

The Severity of Internal Carotid Artery Stenosis is Associated with the Cyclin-Dependent Kinase Inhibitor 2A Gene Expression

open access: yes, 2014
Aim: The INK4b-ARF-INK4a locus in the chromosome 9p21 region is known to play an important role in the development of atherosclerosis. The INK4/ARF transcript p16(INK4a) inhibits the activity of the cyclin-dependent kinases CDK4/CDK6 and arrests cell ...

SIRT6‐Mediated Deacetylation of ATF3 Promotes Silica‐Induced Lung Fibrosis by Enhancing its Nuclear Import via Binding to Importin α

open access: yesAdvanced Science, EarlyView.
SIRT6‐mediated ATF3 acetylation drives MGARP transcription and mitochondrial dysfunction in macrophages, promoting macrophage senescence and pulmonary fibrosis. Mechanistically, HSP70/Importin α competitively binds to ATF3, modulating its nuclear translocation.

Diet and atherosclerosis.

open access: yesSouth African medical journal = Suid-Afrikaanse tydskrif vir geneeskunde, 1974
Among the various factors affecting the development of atherosclerosis and its complications, the diet emerges as an important influence. This article reviews the evidence linking diet and atherosclerosis; the relation between serum cholesterol concentration and incidence of coronary heart disease, and the effect of various dietary components on the ...

Pathological Mechanism‐Inspired Biomimetic Nano‐Senotherapy for Reversing Experimental Atherosclerosis in ApoE−/− Mice

open access: yesAdvanced Science, EarlyView.
The biomimetic self‐assembly nanomedicine reversing atherosclerosis via senotherapy strategy. ABSTRACT The greatest challenge in atherosclerosis (AS) management lies in achieving lesion reversal, not merely slowing progression. Senescent cell accumulation—driven by continuous generation and apoptotic resistance—perpetuates plaque pathology and ...

Circulating long non-coding RNAs as predictors of type 2 diabetes mellitus development: results from the CORDIOPREV study

open access: yesCardiovascular Diabetology
Background Type 2 diabetes mellitus (T2DM) is a growing global health challenge. Conventional diagnostic tools have limited sensitivity and specificity for early-stage disease.

A Dual‐Phase Exosomal Nanotherapy Enhances Continual Efferocytosis by Coordinating Checkpoint Inhibition and Metabolic Reprogramming in Atherosclerosis

open access: yesAdvanced Science, EarlyView.
Here we developed Am@SExo, a dual‐functional engineered exosome that coordinates CD47–SIRPα checkpoint blockade with Arg1 mRNA–mediated metabolic reprogramming. Surface SIRPα promotes recognition of apoptotic cells and plaque targeting, while Arg1 expression enhances arginine–ornithine metabolism, Rac1 activation, and actin remodeling in macrophages ...
