Exploiting macrophage autophagy-lysosomal biogenesis as a therapy for atherosclerosis [PDF]

open access: yes, 2017
Macrophages specialize in removing lipids and debris present in the atherosclerotic plaque. However, plaque progression renders macrophages unable to degrade exogenous atherogenic material and endogenous cargo including dysfunctional proteins and ...

Atg1 kinase organizes autophagosome formation by phosphorylating Atg9 [PDF]

open access: yesAutophagy, 2014
The conserved Ser/Thr kinase Atg1/ULK1 plays a crucial role in the regulation of autophagy. However, only very few Atg1 targets have been identified, impeding elucidation of the mechanisms by which Atg1 regulates autophagy. In our study, we determined the Saccharomyces cerevisiae Atg1 consensus phosphorylation sequence using a peptide array-based ...

TBK1 Induces the Formation of Optineurin Filaments That Condensate with Polyubiquitin and LC3 for Cargo Sequestration

open access: yesAdvanced Science, EarlyView.
Phosphorylation of Optineurin by TBK1 induces the formation of filaments that condensate upon binding to linear polyubiquitin. Membrane‐anchored LC3 partitions into these condensates, suggesting that phase separation of filamentous Optineurin with ubiquitylated cargo promotes the sequestration of cargo and its subsequent alignment with LC3‐positive ...

Alkyltriphenylphosphonium Binding to Cardiolipin Triggers Oncosis in Cancer Cells

open access: yesAdvanced Science, EarlyView.
Alkyltriphenylphosphonium, exemplified by TPP+‐C14, preferentially accumulates in mitochondria and selectively binds to cardiolipin, a key phospholipid of the inner mitochondrial membrane, causing loss of mitochondrial membrane potential, severe cellular ATP depletion, and calcium imbalance.

Intranasal rapamycin ameliorates Alzheimer-like cognitive decline in a mouse model of Down syndrome [PDF]

open access: yes, 2018
Background: Down syndrome (DS) individuals, by the age of 40s, are at increased risk to develop Alzheimer-like dementia, with deposition in brain of senile plaques and neurofibrillary tangles.

Autophagosome Formation by Endophilin Keeps Synapses in Shape [PDF]

open access: yesNeuron, 2016
Soukup et al. (2016), in this issue of Neuron, and Murdoch et al. (2016), in Cell Reports, reveal an unexpected function for the endocytic protein endophilin in autophagosome formation at synapses: preventing neurodegeneration and ataxia.

A Mussel‐Inspired Bioadhesive Patch to Selectively Kill Glioblastoma Cells

open access: yesAdvanced Science, EarlyView.
An innovative mussel‐inspired bioadhesive patch has been developed for post‐surgical glioblastoma treatment. The patch, which adheres strongly in biological environments, releases a localized treatment. This treatment, acting via reactive oxygen species, shows specific toxicity to glioblastoma cells.

Effector Caspase Dcp-1 and IAP Protein Bruce Regulate Starvation-Induced Autophagy during Drosophila Melanogaster Oogenesis [PDF]

open access: yes, 2008
A complex relationship exists between autophagy and apoptosis, but the regulatory mechanisms underlying their interactions are largely unknown. We conducted a systematic study of Drosophila melanogaster cell death–related genes to determine their ...

XIAP Stabilizes DDRGK1 to Promote ER‐Phagy and Protects Against Noise‐Induced Hearing Loss

open access: yesAdvanced Science, EarlyView.
Mechanism of GAS‐mediated protection against noise‐induced hearing loss (NIHL). Noise exposure activates the ATF4/eIF2α axis, downregulating XIAP and promoting DDRGK1 degradation, thereby inhibiting ER‐phagy and leading to hair cell (HC) death. GAS treatment rescues XIAP and DDRGK1 expression, reactivating ER‐phagy to mitigate HC loss, synaptic damage,

ER-Targeted Beclin 1 Supports Autophagosome Biogenesis in the Absence of ULK1 and ULK2 Kinases

open access: yesCells, 2019
Autophagy transports cytoplasmic material and organelles to lysosomes for degradation and recycling. Beclin 1 forms a complex with several other autophagy proteins and functions in the initiation phase of autophagy, but the exact role of Beclin 1 ...
