Bacterial Transformation (Kushner Method)

2003
The principle of this procedure is very similar to that of the standard calcium chloride method, (see Chapter 34 ) i.e., exponential phase cells are harvested and washed with a solution containing divalent cations. This renders the cells competent, which simply means they are now able to take up DNA from the solution.

Mechanism of Bacterial Transformation and Transfection

1974
Publisher Summary This chapter discusses that in spite of the large amount of information on the physiology of the competent cell, including the production of competence factors in a number of transformation systems and an additional protein that must be synthesized to permit irreversible binding of DNA by D.

A simple and rapid method of bacterial transformation

Journal of Microbiological Methods, 2010
Recently, a unique method for bacterial transformation using nanofibers to inoculate DNA has been developed by Naoto Yoshida and colleagues. We have verified the principle, transforming Escherichia coli, Yersinia enterocolitica and Acinetobacter baumannii, and have established a user-friendly protocol. A buffered suspension of sepiolite-an inexpensive,

"Steady-state" competence for bacterial transformation

Canadian Journal of Microbiology, 1968
The cells of Bacillus subtilis, grown in minimal media, are known to become competent for transformation for a short period at a specific phase of ceil growth. In the present work the cells (strain 168 ind−) were grown in continuous culture (chemostat; glucose limiting, generation time 4 h, 37 °C).

DNA uptake during bacterial transformation

Nature Reviews Microbiology, 2004
Naturally competent bacteria are able to take up exogenous DNA and undergo genetic transformation. The transport of DNA from the extracellular milieu into the cytoplasm is a complex process, and requires proteins that are related to those involved in the assembly of type IV pili and type II secretion systems, as well as a DNA translocase complex at the

Current Status of Bacterial Transformations

1955
Publisher Summary When it became clear that a number of plant viruses are crystallizable macromolecules composed of ribonucleic acid and protein, it was possible to attribute to a particular sort of macromolecule the striking biological property of autoreproduction.

Strategies for the development of bacterial transformation systems

Biochimie, 1988
An effective transformation system is a prerequisite for facile genetic manipulation of bacteria. Bacteria may be naturally competent for transformation or may be treated with various agents, such as Tris buffers or divalent metal ions, to induce competence.

Genomic Insights into Bacterial DMSP Transformations

Annual Review of Marine Science, 2012
Genomic and functional genomic methods applied to both model organisms and natural communities have rapidly advanced understanding of bacterial dimethylsulfoniopropionate (DMSP) degradation in the ocean. The genes for the two main pathways in bacterial degradation, routing DMSP to distinctly different biogeochemical fates, have recently been identified.

Bacterial-induced transformation

Science Signaling, 2015
Salmonella infection triggers signaling events that increase the probability of developing cancer in hosts that are genetically predisposed to transformation.

Bacterial Transformations of and Resistances to Heavy Metals

1984
Bacteria carry out chemical transformations of heavy metals. These transformations (including oxidation, reduction, methylation, and demethylation) are sometimes byproducts of normal metabolism and confer no known advantage upon the organism responsible. Sometimes, however, the transformations constitute a mechanism of resistance.
