Regioselective Synthesis of Benzofuranones and Benzofurans
The Journal of Organic Chemistry, 2021Reaction of 3-hydroxy-2-pyrones with nitroalkenes bearing ester groups gives benzofuranones. The reaction allows regioselective preparation of the benzofuranones with programmable substitution at any position. Complex substitution patterns are readily created. The substituted benzofuranones can be converted to substituted benzofurans.

Benzofuran derivatives. Part 3 On the reactivities of the intermediates in benzofuran synthesis
Journal of Heterocyclic Chemistry, 1987Abstract3‐Methyl‐5‐nitrobenzofuran (2) and 3‐methyl‐5‐nitrobenzofuran‐2‐carboxylic acid (3) were obtained by heating 2‐acetyl‐4‐nitrophenoxyacetic acid (1) with various bases in acetic anhydride. It appeared that 3‐hydroxy‐3‐methyl‐5‐nitro‐2,3‐dihydrobenzofuran‐2‐carboxylic acid (4) was the intermediate in the benzofuran synthesis.

Bioorganic & Medicinal Chemistry, 1999Replacement of the 3,4-dialkoxyphenyl substructure common to a number of PDE4 inhibitors with a 2-alkyl-7-methoxybenzofuran unit is described. This substitution can result in either enhancement or substantial reductions in PDE4 inhibitory activity depending on the system to which it is applied.
