Luck or Skill? An Examination of the Ehrlich - Simon Bet [PDF]
In 1980, Paul Ehrlich and Julian Simon placed a famous bet on whether the prices of a bundle of natural resources would rise or fall over the ensuing decade. Simon won the bet as the real price of the bundle fell significantly, and the result of this bet

Finding novel vulnerabilities of hypomorphic BRCA1 alleles
Synthetic lethality screens performed to identify novel vulnerabilities often model complete gene loss, thereby overlooking patient‐derived hypomorphic mutations. In this study, we have performed genome‐wide CRISPR screens on BRCA1 hypomorphic mutations, showing BRCA1I26A behaves like wild‐type, while BRCA1R1699Q mimics deficiency. Furthermore, we have

Dietary compound resveratrol is a pan-BET bromodomain inhibitor
The chemopreventive and anticancer effects of resveratrol (RSV) are widely reported in the literature. Specifically, mechanisms involving epigenetic regulation are promising targets to regulate tumor development. Bromodomains act as epigenetic readers by

Oncogenic DMTF1β promotes cancer cell motility by regulating autophagy through ULK1 stabilization
In the current study, we demonstrate that the oncogene DMTF1β regulates ULK1 stability by reducing its proteasomal degradation in cancer cells. This stabilization enables ULK1 to induce autophagy, which in turn facilitates cancer cell migration. Consequently, reduced DMTF1β levels lead to decreased autophagy and impaired cancer cell migration.

BET isoform selectivity through diverse linkers for bivalent inhibitors : GSK785, a BRD2/4-selective bivalent BET inhibitor [PDF]
The inhibition of the Bromodomain and Extra Terminal (BET) family of proteins has been widely studied for over a decade for its potential therapeutic benefit in cancer and immuno-inflammatory diseases.

Single‐cell multi‐omics reveals epigenetic heterogeneity across therapy‐adaptive tumor states, including quiescent/dormant, drug‐tolerant persister, and EMT‐like phenotypes. By linking regulatory features with state‐associated biomarkers, these approaches inform biomarker‐guided therapeutic strategies for evolving tumors.

Characterizing the Therapeutic Potential of a Potent BET Degrader in Merkel Cell Carcinoma
Studies on the efficacy of small molecule inhibitors in Merkel cell carcinoma (MCC) have been limited and largely inconclusive. In this study, we investigated the therapeutic potential of a potent BET degrader, BETd-246, in the treatment of MCC. We found

I-BET-762 treatment suppresses AHR and inflammatory cells after IFNγ/LPS stimulation.
Mice were administered IFNγ/LPS i.t at 0h. Some mice were treated with I-BET-762 or vehicle (hydroxyl-β cyclodextrin) i.v 1h before and 4h after IFNγ/LPS exposure. Endpoints were assessed at 12h.

Matched spatial transcriptomics and single‐nuclei RNA‐seq were generated for anaplastic and BRAFV600E papillary thyroid cancers revealing generic and tumor‐specific states occurring in cancer cells and in the tumor microenvironment. In this context, cancer dedifferentiation mirrored organoid maturation through ordered thyroid marker gain/loss ...
