Beta-alanine and adaptation in Drosophila

Populations of Drosophila melanogaster show melanistic polymorphism. Adult melanization is inversely related to the rate of incorporation of beta-alanine into tanning cuticles. Light tan pigmentation is directly related to this incorporation. Beta-alanine induced tanning serves to protect pupae from ultraviolet light damage.

The combination of Fourier transform microwave spectroscopy in a pulsed supersonic jet with laser ablation has made beta-alanine amenable to a structural study in the gas phase. Two new conformers of beta-alanine have been identified together with the two previously observed by McGlone and Godfrey [J. Am. Chem. Soc. 1995, 117, 1043].

Starting from porcine insulin, A1-glycine was substituted by beta-alanine. The blood sugar lowering effect of the new analogue in the rabbit is about 45% of that of insulin. The half-maximal binding to partially purified rat liver receptors is about 46%, to transformed human lymphocytes about 54%, compared to insulin.

The synthesis of poly-ß-alanine by a lipase catalyzed polycondensation reaction between ß-alanine esters is reported. The effect of different solvents, reaction temperatures and substrate/enzyme concentrations on polymer yield and degree of polymerization (DP) was determined.
