Cinobufagin Directly Targets PDE4D to Disrupt Fibroblast–Dendritic Cell Crosstalk in Atopic Dermatitis

open access: yesAdvanced Science, EarlyView.
Single‐cell RNA sequencing revealed that MIF signaling drives inflammatory fibroblastmyeloid cell crosstalk in atopic dermatitis. Cinobufagin, a potent PDE4D inhibitor, suppresses MIF and reduces inflammation by restoring the cAMP/PKA/CREB pathway. Genetic knockout validates PDE4D as a key driver and promising therapeutic target for atopic dermatitis ...

Critical Role of IL1R2‐ENO1 Interaction in Inhibiting Glycolysis‐Mediated Pyroptosis for Protection Against Lethal Sepsis

open access: yesAdvanced Science, EarlyView.
The authors have discovered that intracellular interleukin 1 receptor 2 (IL1R2) negatively regulates pyroptosis and inflammation by inhibiting glycolysis in sepsis. Soluble IL1R2 is released from macrophages undergoing pyroptosis. IL1R2 acts as a novel negative regulator of glycolysis by interacting with enolase 1 (ENO1), thereby inhibiting gasdermin D

Dual‐Targeted Nanovesicles Induced Cancer Stem‐Like Cell Differentiation to Sensitize Hepatocellular Carcinoma Radiotherapy

open access: yesAdvanced Science, EarlyView.
The existence of Pin1 and Notch1 crosstalk is identified and verified in HCC, and the crucial function of this crosstalk in sustaining the aggressive phenotypes of CSCs and radioresistance. A biomimetic nanovesicle is now developed to block the crosstalk between Pin1 and Notch1 for inducing CSCs differentiation and sensitizing HCC radiotherapy ...

Ultrasound‐Mediated Biotransfection of Engineered Bone Marrow Mesenchymal Stem Cells in Treated Bone Defects through Intracellular Cavitation

open access: yesAdvanced Science, EarlyView.
Ultrasound‐mediated biotransfection through intracellular cavitation increases plasmid nuclear translocation and gene transfection efficiency by 284.7% and 131.6% in engineered bone marrow mesenchymal stem cells (BMSCs) compared to traditional ultrasound‐targeted microbubble destruction techniques. The maintained bone morphogenetic protein 2 expression
