Antimicrobial Peptide‐Peptoid Macrocycles from the Polymyxin B2 Chemical Space

We optimized mixed peptide‐peptoid sequences for molecular similarity to the macrocyclic lipopeptide antibiotic polymyxin B2 (PMB2) using a genetic algorithm. Synthesis and testing of top‐scoring compounds allowed us to identify new macrocycles with similar antimicrobial activity and reduced toxicity compared to PMB2.

Nitrogen Insertion via Asymmetric Condensation and Chirality Transfer: A Stereodivergent Entry to Cyanocyclopropanes

The synthesis of cyanocyclopropanes is achieved via asymmetric nitrogen insertion into prochiral cyclobutanones. Key mechanistic steps involve an organocatalyzed asymmetric condensation followed by chirality transfer via an intercepted Neber rearrangement.
