Improved Synthesis of the ABCDE Fragment of Brevetoxin A

A second-generation synthesis of the BCDE fragment of brevetoxin A is described. Novel reactions were developed that extend the utility of the asymmetric glycolate alkylation reaction and improve scale-up to provide gram quantities of the B and E subunits. Significant improvements to the convergent assembly of the tetracycle were also realized.

A Highly Convergent Approach to Brevetoxin A

Through optimized, scaled-up routes to the G and J rings of brevetoxin A, a convergent strategy based upon a Horner-Wadsworth-Emmons coupling reaction completed the GHIJ fragment. Further studies toward an alternative protecting group strategy for the GHIJ fragment elucidated a shortened synthesis.

Brevetoxins: unique polyether dinoflagellate toxins

Brevetoxins are lipid‐soluble polyether marine toxins of unique structure and pharmacological function. Toxins are active in vivo in the nanomolar to picomolar concentration range and in vitro in isolated neuromuscular or giant axon preparations and in single‐cell or subcellular model systems.

Distribution and elimination of brevetoxin PbTx-3 in rats

After i.v. administration, [3H]PbTx-3 was rapidly cleared from the blood; less than 10% remained after 1 min. Within 30 min, radiolabel distributed to skeletal muscle (69.5%), liver (18.0%), and intestinal tract (8.0%). Over 24 hr, radiolabel decreased in muscle, remained constant in liver, and increased in the intestinal tract and feces.

Brevetoxins (BTXs) are potent marine toxins produced by certain microalgae during harmful algal blooms (HABs). These lipophilic polyether compounds, notably PbTx-1 and PbTx-2, primarily affect seafood safety by bioaccumulating in filter-feeding shellfish and finfish.
