Nicardipine suppresses bronchoconstrictor actions of pharmacologic agents in guinea pigs

We investigated the inhibitory effects of nicardipine on airway smooth muscle constriction in the guinea pig via both in vitro and in vivo preparations. In the in vitro studies, we created dose-response curves of the excised tracheal strip. The increase in respiratory overflow produced by leukotriene (LT) D4 was measured in the in vivo preparation.

Differences in the response to methacholine between the tidal breathing and dosimeter methods: influence of the dose of bronchoconstrictor agent delivered to the mouth.

It has been postulated that differences in provocative concentration of methacholine causing a 20% fall in FEV1 (PC20) values between the dosimeter method and tidal breathing method might be due to differences in the dose of agonist delivered to the mouth.
