Cancer‐Associated BCL‐2 Mutants Reveal Mechanisms Towards Venetoclax Resistance

open access: yesAdvanced Science, EarlyView.
Venetoclax (VEN) resistance in chronic lymphocytic leukemia arises from diverse BCL2 mutations. We map mechanisms contributing to VEN resistance across common BCL‐2 variants. G101V and D103Y reduce drug binding and increase sequestration of pro‐apoptotic proteins. V156D blocks VEN allosterically.

Caloric Restriction Mimetic 2-Deoxyglucose Alleviated Inflammatory Lung Injury via Suppressing Nuclear Pyruvate Kinase M2–Signal Transducer and Activator of Transcription 3 Pathway

open access: yesFrontiers in Immunology, 2018
Inflammation is an energy-intensive process, and caloric restriction (CR) could provide anti-inflammatory benefits. CR mimetics (CRM), such as the glycolytic inhibitor 2-deoxyglucose (2-DG), mimic the beneficial effects of CR without inducing CR-related ...

Establishing a role for specific nutrients in Drosophila dietary restriction [PDF]

open access: yes, 2009
Dietary restriction (DR), the reduction in food intake that falls short of starvation, has been shown to be the most robust and reproducible intervention to extend lifespan in diverse organisms ranging from yeast to mammals, including the fruit fly ...

Heat Stress Promotes Fibroblast‐Derived WNT5A Secretion Through m6A Modification to Activate Melanogenesis

open access: yesAdvanced Science, EarlyView.
Heat stress promotes WNT5A secretion from fibroblasts via METTL3/YTHDC1‐dependent m6A methylation. Subsequently, WNT5A binds to FZD10 on melanocytes, activating β‐catenin‐driven melanogenesis. ABSTRACT As the impact of global warming continues to intensify, the effects of heat stress on the skin are becoming increasingly evident.

Overcoming Transmission Barriers: A Dual‐Functional Adhesive Hydrogel for Direct Monkeypox Virus Neutralization and Wound Healing

open access: yesAdvanced Science, EarlyView.
MPXV infection leads to persistent viral replication and severe local inflammation, while existing treatments cannot simultaneously remove viruses and repair injured tissue. This study constructs Polymer@mPDA@Gel hydrogel via a two‐step hierarchical assembly method: mPDA anchors photothermal polymers to form antiviral photothermal subunits, which are ...

Trial watch: dietary interventions for cancer therapy

open access: yesOncoImmunology, 2019
Dietary interventions have a profound impact on whole body metabolism, including oncometabolism (the metabolic features allowing cancer cells to proliferate) and immunometabolism (the catabolic and anabolic reactions that regulate immune responses ...

Heat‐Suppressing Projection Two‐Photon Lithography Enables High‐Throughput Sub‐Micrometer Manufacturing of Biopolymer Hydrogels for Tissue Engineering

open access: yesAdvanced Science, EarlyView.
The iLEAP technique integrates high‐throughput two‐photon lithography with extrusion printing, bridging sub‐micrometer precision and macroscopic scaffold fabrication. By employing a low‐exothermic photoinitiator, it mitigates thermal damage to biopolymers and preserves bioactivity.
