The Role of the Casein Kinase 1 (CK1) Family in Different Signaling Pathways Linked to Cancer Development

open access: yesOncology Research and Treatment, 2005
The members of the casein kinase 1 (CK1) family are highly conserved and are expressed in many eukaryotes ranging from yeast to humans. Mammalian CK1 isoforms (alpha, beta, gamma, delta, epsilon) and their splice variants are involved in diverse cellular processes including membrane trafficking, circadian rhythm, cell cycle progression, chromosome ...

Identification and Profiling of a Selective and Brain Penetrant Radioligand for in Vivo Target Occupancy Measurement of Casein Kinase 1 (CK1) Inhibitors

ACS Chemical Neuroscience, 2017
To enable the clinical development of our CNS casein kinase 1 delta/epsilon (CK1δ/ε) inhibitor project, we investigated the possibility of developing a CNS positron emission tomography (PET) radioligand. For this effort, we focused our design and synthesis efforts on the initial CK1δ/ε inhibitor HTS hits with the goal of identifying a compound that ...

Gene expression levels of Casein kinase 1 (CK1) isoforms are correlated to adiponectin levels in adipose tissue of morbid obese patients and site-specific phosphorylation mediated by CK1 influences multimerization of adiponectin

Molecular and Cellular Endocrinology, 2015
White adipose tissue has now been recognized as an important endocrine organ secreting bioactive molecules termed adipocytokines. In obesity, anti-inflammatory adipocytokines like adiponectin are decreased while pro-inflammatory factors are over-produced. These changes contribute to the development of insulin resistance and obesity-associated diseases.

The role of the casein kinase 1 (CK1) family in fungal infections

Molecular Biology Reports
The Casein kinase 1 (CK1) family, as an important member of the protein kinase family, exhibits high evolutionary conservation and is widely distributed in various cell types. CK1 plays critical roles in numerous cellular biological processes, and its abnormal expression is closely associated with the development and progression of various diseases ...

Discovery of Inhibitor of Wnt Production 2 (IWP-2) and Related Compounds As Selective ATP-Competitive Inhibitors of Casein Kinase 1 (CK1) δ/ε

Journal of Medicinal Chemistry, 2018
Inhibitors of Wnt production (IWPs) are known antagonists of the Wnt pathway, targeting the membrane-bound O-acyltransferase porcupine (Porcn) and thus preventing a crucial Wnt ligand palmitoylation. Since IWPs show structural similarities to benzimidazole-based CK1 inhibitors, we hypothesized that IWPs could also inhibit CK1 isoforms.

Abstract 604: Targeting casein kinase 1 alpha (CK1 alpha) and transcriptional CDKs (CDK7/9) in human liposarcomas

Cancer Research
Abstract Introduction: Liposarcomas (LPS) are rare mesenchymal cell malignancies of adipocytic origin that are diagnosed in more than 3500 patients in the US each year. The two most common subtypes, well-differentiated LPS (WDLPS) and dedifferentiated LPS (DDLPS), are characterized by extrachromosomal DNA amplifications harboring the ...

Abstract 2980: Therapeutic potential of combined targeting of casein kinase 1 alpha (CK1 alpha) and CDK7/9 with the inhibitor BTX-A51 in human liposarcomas

Cancer Research
Abstract Introduction: Liposarcomas (LPS) are mesenchymal cell malignancies that are diagnosed in more than 3500 patients in the US each year. LPS are characterized by DNA amplifications of MDM2 (100%) and CDK4 (90+%) genes on neochromosomes, generally with wild-type TP53.
