[Integrase inhibitors and CCR5 receptor antagonists].

Integrase inhibitors and CCR5 receptor antagonists, novel class of antiretrovirals, were introduced into clinical practice in late 2000s. Currently available drugs in these classes, raltegravir and maraviroc, show good tolerability and have little impact on lipid or glucose metabolism compared to traditional classes.

Inhibition of HIV infection by CXCR4 and CCR5 chemokine receptor antagonists.

The chemokine receptors CXCR4 and CCR5 are used as co-receptors by the T cell-tropic (X4) and macrophage-tropic (R5) HIV-1 strains, respectively, for entering their host cells. Viral entry can be inhibited by the natural ligands for CXCR4, the CXC chemokine SDF-1 and CCR5, the CC chemokines RANTES, MIP-1alpha and MIP-1beta.

Pfizer Inc is developing maraviroc, a CCR5 receptor antagonist for the treatment of HIV-1 infection and rheumatoid arthritis. In April 2007, the FDA advisory committee voted to approve maraviroc for HIV-1 infection. Phase II and III clinical trials, and post-approval studies are ongoing in both treatment-experienced and -naive HIV patients.
