Gardiquimod Nanoemulsion Targets Cutaneous Leishmaniasis Lesions Reducing Systemic Toxicity and Parasite Burden

open access: yesAdvanced Healthcare Materials, EarlyView.
Nanoemulsion delivery of a TLR7 agonist enhances safety and promotes colocalization with Leishmania‐infected macrophages in skin lesions, enabling a ∼2‐log reduction in parasite burden. However, tissue‐level constraints and TLR‐driven regulatory feedback generate a mixed immune response that limits complete parasite clearance.

CD8+ Trms against malaria liver-stage: prospects and challenges

open access: yesFrontiers in Immunology
Attenuated sporozoites provide a valuable model for exploring protective immunity against the malarial liver stage, guiding the design of highly efficient vaccines to prevent malaria infection.

Temporal Immune and Metabolic Shifts Drive the Anti‐Tumor Efficacy of Resiquimod‐Loaded Nanoparticles in Peritoneal Carcinomatosis

open access: yesAdvanced Healthcare Materials, EarlyView.
Peritoneal carcinomatosis is a hard‐to‐treat spread of abdominal cancers that often returns after therapy. Nanoparticles were developed to retain an immune‐boosting drug in the abdomen. When combined with chemotherapy in mice, this approach eliminated tumors in many cases and prevented recurrence, helping the immune system mount a stronger, longer ...

Antigen-specific decidual CD8+ T cells include distinct effector memory and tissue-resident memory cells

open access: yesJCI Insight, 2023
Maternal decidual CD8+ T cells must integrate the antithetical demands of providing immunity to infection while maintaining immune tolerance for fetal and placental antigens.

Encoded Cell‐Material Interactions to Reroute Cytokine Signaling for Regenerative Medicine

open access: yesAdvanced Healthcare Materials, EarlyView.
We present native MATRIX (Material Activated To Regulate Inducible gene eXpression), a co‐engineered material‐cell platform. Surfaces functionalized with antibodies for endogenous soluble ligands (e.g., IL‐1β or IL‐6) capture and immobilize ligands for presentation to a cognate, engineered synthetic Notch receptor, releasing a transcription factor (TF)

Common gamma chain cytokines promote rapid in vitro expansion of allo-specific human CD8+ suppressor T cells. [PDF]

open access: yesPLoS ONE, 2011
Human CD8(+) regulatory T cells, particularly the CD8(+)CD28(-) T suppressor cells, have emerged as an important modulator of alloimmunity. Understanding the conditions under which these cells are induced and/or expanded would greatly facilitate their ...

Assessing Cd8 T Cell Number and Dysfunction in the Presence of Antigen [PDF]

open access: yesThe Journal of Experimental Medicine, 2001
CD8 T cells proliferate and differentiate into cytokine-secreting cytolytic effectors on encounter with viral antigens. As virus is cleared the activated T cells undergo apoptosis, but some survive and enter the memory pool where they persist indefinitely, slowly dividing in a relatively quiescent state until reactivation on reexposure to the antigen 1.

MHC-based detection of antigen-specific CD8+ T cell responses [PDF]

open access: yesCancer Immunology, Immunotherapy, 2010
The hallmark of adaptive immunity is its ability to recognise a wide range of antigens and technologies that capture this diversity are therefore of substantial interest. New methods have recently been developed that allow the parallel analysis of T cell reactivity against vast numbers of different epitopes in limited biological material.

Nanomaterial Strategies for Pulmonary Delivery of Immunotherapeutics in Lung Cancer Treatment

open access: yesAdvanced Healthcare Materials, EarlyView.
Inhalable immunotherapeutic nanomedicines enable organ‐selective immune modulation by overcoming pulmonary delivery barriers and concentrating therapy within lung tumors. This Review defines how nanomaterial properties govern airway deposition, retention, cellular partitioning, and immune activation across vaccines, checkpoint blockade, STING agonists,

Turning Unpredictable Biomolecule Adsorption to Controlled Corona Formation: Focus on Carbon Nanomaterials

open access: yesAdvanced Materials, EarlyView.
Controlling the protein corona formation onto carbon nanomaterials (CNMs) enhances their functionalities as platforms for cancer theranostics. Here, we reviewed the effects of the intrinsic and acquired properties of CNMs on protein corona formation, the consequent biological and toxicological outcomes, and the strategies to reshape corona formation ...
