Digital microfluidics for cell-based assays

Lab on a Chip, 2008
We introduce a new method for implementing cell-based assays. The method is based on digital microfluidics (DMF) which is used to actuate nanolitre droplets of reagents and cells on a planar array of electrodes. We demonstrate that this method is advantageous for cell-based assays because of automated manipulation of multiple reagents in addition to ...

Cell-based assays: fuelling drug discovery

Analytical and Bioanalytical Chemistry, 2010
It has been estimated that over a billion dollars in resources can be consumed to obtain clinical approval, and only a few new chemical entities are approved by the US Food and Drug Administration (FDA) each year. Therefore it is of utmost importance to obtain the maximum amount of information about biological activity, toxicological profile ...

Cell-Based Bioluminescence Screening Assays

2013
Drug screening is an essential and widely used technique for drug discovery in various biomedical fields notably in oncology. Here we describe a functional screening assay based on the bioluminescence detection of a secreted luciferase for monitoring cell viability of cancer cells in a high-throughput format.

Cell-Based Assays

2005
This chapter describes methods and tools for processing and visualizing data from high-throughput cell-based assays. Such assays are used to examine the contribution of genes to a biological process or phenotype (Carpenter and Sabatini, 2004). In principle, this can be done for any gene or combination of genes and for any biological process of interest.

Cell Based Biological Assay Using Microfluidics

2008
A microfluidic device has been designed to measure and manipulate microdroplets, in which protein expression is induced in single cells. The device exploits the permeation of water through poly (dimethylsiloxane) (PDMS) in order to keep the concentration of solutes in aqueous picoliter volume microdroplets stored in wells.

Label-Free Cell-Based Functional Assays

Combinatorial Chemistry & High Throughput Screening, 2008
Label-free technologies based on electrical impedance or refractive index are new tools for measuring a cell-based functional response. Although the technologies are relatively new to high throughput screening cell-based applications, they are rapidly generating interest in that they are able to measure a phenotypic response using cells natively ...

Cell-based assays for kinase drug discovery

Drug Discovery Today: Technologies, 2010
Kinases are important therapeutic targets. Potent in vitro inhibitors are readily produced, but these compounds are not always potent in cells. Because cell potency is crucial for efficacy, cell-based assays are important for successful inhibitor development. In this review kinase cell assays are divided into four types.

Cell-based assays on microfluidic chips

2023
In view of miniaturization, different microfluidic devices have been designed and developed to overcome the difficulties of conventional cell-based assays in terms of throughput, single-cell isolation, cost, and flexibility. These platforms are advantageous because of their low reagent consumption, small amount of sample required, as well as facile ...

Cell-Based Assays for High-Throughput Screening

Molecular Biotechnology, 2010
Cell-based assays represent approximately half of all high-throughput screens currently performed. Here, we review in brief the history and status of high-throughput screening (HTS), and summarize some of the challenges and benefits associated with the use of cell-based assays in HTS.

Cell based assays

2017
Tumor necrosis factor alpha (TNFa) not only plays a key role in the human immune system, but also has a cytotoxic effect on tumor cells. Owing to this function, TNFa can become a considerable candidate for chemotherapy but needs to be delivered locally.
