A computational pipeline to quantify perinuclear lysosomes in fibroblasts using CellProfiler v1

open access: yes, 2023
Here we present a CellProfiler software pipeline to quantify the distribution of lysosomes in MEF cells. The lysosomes were stained using anti-LAMP1 antibody, and nuclei were labeled using DAPI. The images were acquired using a Zeiss laser scanning confocal microscope and were maximum intensity projected in FIJI.

Gated Spatial Hidden Markov Models for Classification in Histological Samples, With Application to Muscle Fiber‐Typing

open access: yesStatistics in Medicine, Volume 45, Issue 23-24, October 2026.
ABSTRACT Quantitative histology often requires assigning each site or cell to one of two classes from continuous stain intensities. In practice, large‐scale illumination and contrast fluctuations bias thresholds and obscure true biological differences.

Autophagy Flux Is Remodeled Sex‐ and Cell Type‐Specifically During Human Aging, and Is Linked to Reduced Physical Function in Older Adults

open access: yesAging Cell, Volume 25, Issue 10, October 2026.
Autophagy‐gene transcription increases with age and is uncoupled from autophagy flux. Autophagy flux is remodeled sex‐ and cell type‐specifically and is associated with worse physical function in older adults. In a pilot intervention study, elevated PBMC autophagy flux decreased following mild exercise in older adults.

Computer Vision for Substrate Detection in High‐Throughput Biomaterial Screens Using Bright‐Field Microscopy

open access: yesAdvanced Intelligent Systems
High‐throughput screening (HTS) can be used when ab initio information is unavailable for rational design of new materials, generating data on properties such as chemistry and topography that control cell behavior.

Protracted Fate Acquisition and Epigenetic De‐Aging During Induced Neural Stem Cell Conversion of Human Blood Cells

open access: yesAging Cell, Volume 25, Issue 10, October 2026.
In contrast to rapid epigenetic de‐aging during induced pluripotent stem cell generation, direct conversion of blood cells into neural stem cells is associated with largely protracted epigenetic de‐aging. The process coincides with prolonged cell fate consolidation, does not require sustained proliferation, and might provide experimental access to ...

Reln Haploinsufficiency Enhances Fentanyl‐Induced Locomotion and Dorsal Striatal Activity Without Affecting Opioid Reinforcement and Relapse‐Like Behavior

open access: yesGenes, Brain and Behavior, Volume 25, Issue 5, October 2026.
Reln haploinsufficiency does not alter fentanyl self‐administration, relapse‐like seeking, or conditioned place preference in mice. Instead, reduced Reln expression selectively enhances acute fentanyl‐induced locomotion and dorsal striatal Fos activation, positioning Reelin as a modulator of opioid‐evoked neuronal activity rather than opioid ...

Modular training resources for bioimage analysis

open access: yesJournal of Microscopy, Volume 304, Issue 1, Page 3-14, October 2026.
Abstract Modern microscopy enables us to measure structural and dynamical properties of many biological processes and is therefore an indispensable research tool. However, the amount and complexity of the produced imaging data is steadily increasing.

Challenging specificity of chemicalcompounds targeting GPCRs with cellprofiling

open access: yes, 2020
Screening compounds with image-based analysis is an important part in the processof drug discovery. It is an efficient way to screen compounds as it gives moreinformation than for example HTS. High-content screening as it is also called, hasreally progressed in recent years, as the field of data science evolves, and with it sodoes the efficiency of how

Macroscopic to ultrastructural analyses identify the loss of myofibrils as the primary mediator of ageing‐ and disuse‐induced muscle fibre atrophy

open access: yesThe Journal of Physiology, Volume 604, Issue 20, Page 8523-8541, 15 October 2026.
Abstract figure legend Skeletal muscle structure was analysed at three levels in young vs. aged humans, and in young vs. aged mice subjected to unilateral hindlimb immobilization to model disuse. (1) Macroscopic: magnetic resonance imaging of the thigh (human) and wet mass of individual hindlimb muscles (mouse) revealed that both ageing and disuse ...
