Protonophore activity of short‐chain fatty acids induces their intracellular accumulation and acidification

open access: yesFEBS Letters, EarlyView.
The protonated form of butyrate, as well as other short‐chain fatty acids (SCFAs), is membrane permeable. In acidic extracellular environments, this can lead to intracellular accumulation of SCFAs and cytosolic acidification. This phenomenon will be particularly relevant in acidic environments such as the large intestine or tumor microenvironments ...

ASIC currents in cultured primate retinal amacrine/ganglion cells

open access: yesPhysiological Reports
Acid‐sensing ion channels (ASICs) are proton‐gated cation channels belonging to the epithelial Na + channel/degenerin superfamily. In the CNS, ASICs are involved in synaptic plasticity, learning/memory, and acidosis‐mediated injury.

Modifications in FLAP's second cytosolic loop influence 5‐LOX interaction, inhibitor binding, and leukotriene formation

open access: yesFEBS Letters, EarlyView.
The enzyme 5‐lipoxygenase (5‐LOX) catalyzes the first step in the biosynthesis of leukotrienes (LTs) involved in inflammatory pathophysiology. After cellular stimulation, 5‐LOX translocates to the nucleus, interacting with the 5‐LOX‐activating protein (FLAP) to form LTA4 from arachidonic acid (AA).

Brucella NyxA and NyxB dimerization enhances effector function during infection

open access: yesFEBS Letters, EarlyView.
Brucella abortus thrives inside cells thanks to the translocation of effector proteins that fine‐tune cellular functions. NyxA and NyxB are two effectors that destabilize the nucleolar localization of their host target, SENP3. We show that the Nyx proteins directly interact with each other and that their dimerization is essential for their function ...

Evaluation of tissue reaction, cell viability and cytokine production induced by Sealapex Plus

open access: yesJournal of Applied Oral Science, 2011
OBJECTIVE: The aim of this study was to investigate the effects of mineral trioxide aggregate (MTA), Sealapex, and a combination of Sealapex and MTA (Sealapex Plus) on the reaction of subcutaneous connective tissue of rats, and on cell viability and ...

Extraction and purification of tumorigenic Schwann cells in vitro from neurofibromatosis type 2 tissues

open access: yesChinese Journal of Contemporary Neurology and Neurosurgery, 2011
Objective To establish a simple method to extract and purify tumorigenic Schwann cells (TSCs) from neurofibromatosis type 2 (NF2) tumor tissues in vitro, in order to improve the successful rate of primary culture and obtain a large number of TSCs from ...

Cell culture

open access: yes, 1996
This chapter provides the tools for using cell culture as an adjunct to the cell biology and molecular biology methods that break down the traditional borders between the disciplines of biology and virology. Cell culture allows looking at cells in their entirety from the outside before examining the intricacy of their component parts. Cells are counted

Platelet lysate as a serum substitute for 2D static and 3D perfusion culture of stromal vascular fraction cells from human adipose tissue [PDF]

open access: yes, 2009
Fetal bovine serum (FBS) and fibroblast growth factor (FGF)-2 are key supplements for the culture of stromal vascular fraction (SVF) cells from adipose tissue, both for typical monolayer (2D) expansion and for streamlined generation of osteogenic ...

The carboxylate “gripper” of the substrate is critical for C‐4 stereo‐inversion by UDP‐glucuronic acid 4‐epimerase

open access: yesFEBS Letters, EarlyView.
UDP‐glucuronic acid 4‐epimerase (UGAepi) catalyzes NAD+‐dependent interconversion of UDP‐glucuronic acid (UDP‐GlcA) and UDP‐galacturonic acid (UDP‐GalA) via C4‐oxidation, 4‐keto‐intermediate rotation, and C4‐reduction. Here, Borg et al. examined the role of the substrate's carboxylate group in the enzymic mechanism by analyzing NADH‐dependent reduction

In vitro study on the mechanisms of action of a novel phytotherapeutic compound against human hepatoma cells

open access: yesAnnals of Hepatology, 2007
HepG2 human hepatoma cells were incubated for 24 or 48 h with various concentrations of YHK solution. After 24 h incubation, cell proliferation and cytotoxicity were determined by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl ...
