Animal stroke models and outcome evaluation: A review

open access: yesAnimal Models and Experimental Medicine, EarlyView.
Rodents are the preeminent stroke model species. Large animal models exist too, but are poorly standardized. Ischemic and hemorrhagic experimental models are available, with the intraluminal filament model being predominant. Outcomes are evaluated via quantifying subjects' sensorimotor impairment, cognitive memory faculties, degree of cellular ...

Animal models for investigating post‐stroke cognitive impairment: A neurovascular trafficking perspective

open access: yesAnimal Models and Experimental Medicine, EarlyView.
Animal models enable mechanistic testing of how ischemic injury disrupts neurovascular trafficking and drives cognitive vulnerability after stroke. Complementary models capture distinct vascular, cellular, white matter, and cognitive features of PSCI.

Cerebrospinal Fluid Cytokine Expression and Relationships With Neurological Measures in Virologically Controlled People With HIV

open access: yesAnnals of Neurology, EarlyView.
Objective Quantify cytokines in cerebrospinal fluid (CSF) of virologically suppressed people with HIV (PWH) and people without HIV (PWoH) and correlate these with neuroimaging, neuropsychometric (NP), and neurodegenerative measurements. Methods CSF samples were collected from virologically suppressed PWH and demographically matched PWoH under a natural

Hyperostosis frontalis interna: Observed patterns, prevalence, and dementia associations in older adults

open access: yesThe Anatomical Record, EarlyView.
Abstract Hyperostosis frontalis interna (HFI) is a frequently underrecognized condition characterized by overgrowth of the frontal endocranium, most commonly observed in post‐menopausal women. Although its etiology remains unclear, hormonal influences and a possible relationship with neurocognitive changes have been proposed.

Diabetes Mellitus Is Associated With Increased Risk for Subsequent Hip Arthroscopy After Index Hip Arthroscopy

open access: yesArthroscopy, Sports Medicine, and Rehabilitation, EarlyView.
Purpose To determine whether diabetes mellitus is associated with higher systemic and hip‐specific complications and to evaluate reoperation rates after hip arthroscopy. Methods Using the TriNetX Research Network and Current Procedural Terminology and International Classification of Diseases codes, we identified diabetic and nondiabetic patients aged ...

Sustained Inhibition of Structural Damage Progression and Improvements in Disease Activity With Guselkumab in Biologic‐Naïve Participants with Active and Erosive Psoriatic Arthritis: Results Through Week 48 of the Phase 3b Randomized, Double‐Blind, Placebo‐Controlled APEX Study

open access: yesArthritis &Rheumatology, Accepted Article.
Objective In APEX, guselkumab, which binds to the IL‐23p19 subunit and CD64 on IL‐23‐producing cells, demonstrated significant joint efficacy and inhibition of structural damage progression in participants with active and erosive PsA through Week (W)24; W48 findings are reported herein. Methods The APEX modified full analysis set included 1020 biologic‐

Efficacy, safety and cost‐effectiveness of CAR‐T therapy

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
CAR T‐cells demonstrate high efficacy in blood cancers, including ALL, MM and DLBCL. Innovations target solid tumours despite challenges such as antigen escape. Combination therapies enhance the delivery and infiltration of CAR T cells. Toxicity, cost and resistance remain major barriers to clinical use.

The integrated Lei‐CNS PBPK‐PD model effectively predicts clozapine brain disposition and D2 receptor occupancy in humans

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Aim Clozapine, used for treatment‐resistant schizophrenia, has a narrow therapeutic window and highly variable plasma pharmacokinetics (PK), necessitating careful monitoring. Direct measurement of unbound brain PK in humans is unethical. To develop a physiologically based pharmacokinetic–pharmacodynamic (PBPK‐PD) model to predict clozapine brain ...

Histamine and the mechanisms of nausea and vomiting: Translating the uncertain pharmacology of cyclizine

open access: yesBritish Journal of Clinical Pharmacology, EarlyView.
Abstract Cyclizine is a long‐established, short‐acting drug, registered for preventing motion sickness, post‐operative nausea and vomiting (PONV) and ‘vomiting of known cause’. The latter is not defined but may include pregnancy and patients receiving palliative care, historically accepted without large, controlled trials.

Cerebral Metabolic Crisis in Pediatric Cerebral Malaria

Journal of Pediatric Intensive Care, 2021
AbstractCerebral metabolic energy crisis (CMEC), often defined as a cerebrospinal fluid (CSF) lactate: pyruvate ratio (LPR) >40, occurs in various diseases and is associated with poor neurologic outcomes. Cerebral malaria (CM) causes significant mortality and neurodisability in children worldwide.
