Production of Cyclodextrins by CGTase from Bacillus clausii Using Different Starches as Substrates

Cyclodextrins (CDs) are cyclic oligasaccharides composed by D-glucose monomers joined by alpha-1,4-D glicosidic linkages. The main types of CDs are alpha-, beta- and gamma-CDs consisting of cycles of six, seven, and eight glucose monomers, respectively.

Molecular cloning and characterization of a novel γ-CGTase from alkalophilic Bacillus sp.

We found a novel cyclodextrin glucanotransferase (CGTase) from alkalophilic Bacillus sp. G-825-6. The enzyme was expressed in the culture broth by recombinant Bacillus subtilis KN2 and was purified and characterized. Isolation and Characterization of A B. Circulans Strain and Its CGTase

A CGTase-secreting bacterial strain was isolated from soil and bacteriologically identified to be Bacillus circulans . Its enzyme was found to be a β-CD type CGTase. The production of this enzyme was undertaken in a 2L-fermentor following medium optimization in terms of carbon and nitrogen sources.

B1 by directed evolution.A library of CGTase mutants was constructed by introducing random mutagenesis using error-prone PCR to screen mutant enzymes with improved extracellular enzyme activities and soluble expression.
