Cryo‐electron microscopy structures reveal two distinct neutralization mechanisms of fully human monoclonal antibodies targeting the Shiga toxin 1a B subunit. RDS059 occludes the Gb3 receptor‐binding site, whereas RDS045 engages a lateral quaternary epitope to disrupt holotoxin assembly.

AI‑guided single‑cell transcriptomics identifies OGT as a key regulator enriched in rheumatoid arthritis fibroblast‑like synoviocytes (RA‐FLSs). Inflammatory stimuli activate the OGT–SAP130 axis, which modulates the H3K27ac/H3K27me3 balance and epigenetically silences BTG2, promoting aggressive transformation of RA‑FLSs.

Chimeric Antigen Receptor Therapy for B-cell Malignancies
We presented data showing that the CART-19 cells expressing the 4-1BB signaling domain can have unprecedented and massive in-vivo expansion, traffic to tumor sites, persist long term in vivo, and induce rapid and potent anti-tumor activity in ...

CAR‐Engineered Cell Therapies Beyond Cancer: Reprogramming Fibrosis and Immune‐Mediated Inflammation
CAR‐engineered cell therapies are expanding beyond cancer toward immune resetting, pathological‐cell clearance, matrix remodeling, and microenvironmental reprogramming in autoimmune, inflammatory, and fibrotic diseases. This Review compares CAR‐T, CAR‐macrophage, and CAR‐NK platforms and proposes controllable spatiotemporal reprogramming to align ...

A new class of lysosome‐directed molecular glue degraders selectively enhance CAPRIN1–APP interactions, driving APP degradation and reducing amyloid‐β production in human neurons and Alzheimer's disease mouse models. This CAPRIN1‐dependent targeted protein degradation strategy reveals a previously unrecognized therapeutic approach for disrupting the ...

ABSTRACT Cardiovascular adverse events (CVAEs) associated with bispecific T‐cell engaging antibodies (BsAbs) have not been systematically investigated across approved agents. In this disproportionality analysis of FAERS (December 2014–September 2025), reports listing BsAbs as the primary suspected drug (n = 7647) were compared with all other drugs in ...

ABSTRACT Current treatments for immune thrombocytopenia (ITP) and warm autoimmune hemolytic anemia (wAIHA), rare autoimmune diseases in which autoreactive B cells play a major role, can lead to high response rates; however, for many patients these responses are not durable or maintained after treatment discontinuation.

Application of adoptive cell therapy in malignant melanoma
Cutaneous melanoma is one of the most aggressive skin cancers originating from skin pigment cells. Patients with advanced melanoma suffer a poor prognosis and generally cannot benefit well from surgical resection and chemo/target therapy due to ...

Confronting the Analytical Challenges of Chimeric Antigen Receptor T Cell [PDF]
Intensive research and development effort has been devoted to Chimeric antigen receptor T (CAR-T) cell therapies. CAR-T presents unique challenges for analytical development due to its complex mechanism of actions, large heterogeneity of the starting ...
