Metformin Restores Mitochondrial Function and Neurogenesis in POLG Patient‐Derived Brain Organoids
Advanced Science, EarlyView.Patient‐derived POLG‐mutant cortical organoids reveal neuronal subtype‐specific mitochondrial and synaptic defects, with dopaminergic neurons most affected. Metformin treatment restores neuronal identity, mitochondrial function, and excitability, increased mtDNA maintenance, and reprogrammed metabolism via TCA and redox pathways.Zhuoyuan Zhang, Tsering Yangzom, Ning Lu, Shenglong Deng, Xianglu Xiao, Guang Yang, Kristina Xiao Liang +6 morewiley +1 more sourcePotentiating the antitumour response of CD8+ T cells by modulating cholesterol metabolism

Advanced Science, EarlyView. This study identifies the valvular interstitial cell populations responsible for valvular calcification induced by hyperphosphatemia and likely aging, uncovers local vitamin D metabolism defect‐induced inflammation as a critical pathogenic factor of calcific aortic valve disease, and highlights active vitamin D and ERK inhibitor as potential preventive Ruichen Yang, Chong Han, Yangli Xie, Shoutao Qiu, Shaoyang Zhang, Jingjia He, Zejian Wang, Zhenlin Zhang, Huijuan Liu, Lin Chen, Baojie Li +10 morewiley +1 more sourceAllosteric Modulation of Pathological Ataxin‐3 Aggregation: A Path to Spinocerebellar Ataxia Type‐3 Therapies
Advanced Science, EarlyView. This study uncovers a new allosteric site in the Josephin domain of ataxin‐3 targeted by the molecular tweezer CLR01, which modulates protein aggregation, improves synaptic function in neuronal cells, and delays motor dysfunction in animal models.Alexandra Silva, Sara Duarte‐Silva, Pedro M. Advanced Science, EarlyView. The study demonstrates that endothelial cell (EC)‐derived BMP6 promotes the osteogenic differentiation of vascular smooth muscle cells (VSMCs) through cell‐cell interactions. Additionally, the researchers preliminarily explore the driving effect of hemodynamic factors on BMP6‐induced calcification and reveal the regulatory role of KLF4 on BMP6.Shen Li, Shuang Cao, Peipei Li, Feng Zhang, Gangfeng Ren, Xin Wang, Jiawei Zhao, Chen Liu, Yuan Gao, Jie Xu, Yongjun Wang, Zongping Xia, Yuming Xu +12 morewiley +1 more source
