Genetic variation in the cholesterol 24-hydroxylase (CYP46) gene and the risk of Alzheimer's disease

Neuroscience Letters, 2002
Alzheimer's disease (AD) is a complex, multifactorial disorder, with many genetic and environmental factors implicated in disease onset and pathology. Increasing evidence points to a link between brain cholesterol turnover and AD. The CYP46 gene encodes for the enzyme, cholesterol 24-hydroxylase, which plays a key role in brain cholesterol turnover.

Lack of association of the cholesterol 24-hydroxylase (CYP46) intron 2 polymorphism with Alzheimer’s disease

Neuroscience Letters, 2004
An association was recently reported between an increased risk of Alzheimer's disease and an intron 2 AA genotype of CYP46, the enzyme hydroxylating cholesterol to 24S-hydroxycholesterol. Moreover, CYP46 AA-carriers were found to have increased levels of amyloid-beta and tau in brain and cerebrospinal fluid. We determined the CYP46 intron 2 genotype in

High Cholesterol Diet Increases Expression of Cholesterol 24-Hydroxylase and BACE1 in Rat Hippocampi: Implications for the Effect of Diet Cholesterol on Memory

Iranian Red Crescent Medical Journal, 2016
Background: Abnormal cholesterol homeostasis is associated with the pathogenesis of neurodegenerative disease and cognitive impairment. Objectives: Our objective was to evaluate changes in the expression of proteins related to cognition and cholesterol homeostasis in the hippocampi of rats as well as behavioral modifications following the ...

Imidazo[1,2-a]pyridines as cholesterol 24-hydroxylase (CYP46A1) inhibitors: a patent evaluation (WO2014061676)

Expert Opinion on Therapeutic Patents, 2014
A series of imidazo[1,2-a]pyridine analogs that effectively inhibit cholesterol 24-hydroxylase (CYP46A1) was reported in WO2014061676. The treatment or prophylaxis of neurodegenerative disorders, such as Alzheimer's disease and Parkinson's disease, by these novel CYP46A1 inhibitors is claimed in the patent.

Cholesterol homeostasis in the nervous system and retinal pathologies: Role of cholesterol 24S-hydroxylase (CYP46A1)

2014

Radiosynthesis and Evaluation of Novel Cholesterol 24-Hydroxylase Positron Emission Tomography Tracers

ABSTRACT Cholesterol 24-hydroxylase (CH24H or CYP46A1) is a pivotal enzyme in brain cholesterol metabolism and has emerged as a therapeutic and imaging target in neurodegenerative disorders. Although [ 18 F]Cholestify ([ 18 F]CHL-2205) has shown ...

In vivo consequences of cholesterol-24s-hydroxylase (CYP46A1) inhibition by voriconazole on cholesterol homeostasis and function in the rat retina

2011
Cholesterol 24S-hydroxylase (CYP46A1) converts cholesterol into 24S-hydroxycholesterol in neurons and participates in cholesterol homeostasis in the central nervous system, including the retina. We aimed to evaluate the consequences of CYP46A1 inhibition by voriconazole on cholesterol homeostasis and function in the retina.
