[249] Choline acetyltransferase
1971Publisher Summary This chapter discusses the methods of preparation of Choline Acetyltransferase. Rates of synthesis of acetylcholine are linear with choline acetyltransferase concentration if all other components of the reaction mixture are added in excess.

Molecular biology and neurobiology of choline acetyltransferase
Molecular Neurobiology, 1987In the 45 years since the first description of choline acetyltransferase (ChAT; EC 2.3.1.6.), significant progress has been made in characterizing the molecular properties of this important neurotransmitter synthetic enzyme. We are now on the verge of understanding its genetic regulation and biological function(s).

Biochemistry and Immunocytochemistry of Choline Acetyltransferase
1986Choline acetyltransferase (ChAT, acetyl CoA-choline-O-acetyltransferase, EC 2.3.1.6) is the enzyme responsible for catalyzing the synthesis of the important central and peripheral nervous system neurotransmitter, acetylcholine (ACh), and therefore the regulation of ChAT activity is likely to be a controlling factor in availability of ACh for neural ...

Choline Acetyltransferase and Neuronal Maturation
1973Publisher Summary This chapter mentions the development of choline acetyltransferase in a restricted segment of the developing nervous system in relation to other parameters of neuronal maturation. A number of studies have indicated that the use of a small segment of the developing neural axis can reveal pronounced changes in the biochemical ...

Interaction of analogs of coenzyme A with choline acetyltransferase
Biochemistry, 1977The finding that methyl methanethiolsulfonate appears to inhibit choline acetyltransferase from squid ganglia not by reacting with a thiol group of the enzyme but by reacting with the thiol group of coenzyme A to form a competitive inhibitor of acetyl coenzyme A led to the synthesis of the ethyl, propyl, and 3-carboxy-4-nitrophenyl disulfides of CoA ...

Inhibition of brain choline acetyltransferase by tetraplatin
Brain Research Bulletin, 1987Tetraplatin inhibited choline acetyltransferase (CAT) from rat and bovine brain with I50 values of 13.5 and 40 microM, respectively. At 0.5 mM concentration, tetraplatin exhibited inhibition of the rat brain enzyme by 89%, whereas cisplatin yielded only 18% inhibition.

Purification of chicken brain choline acetyltransferase
Neurochemistry International, 1981Chicken brain choline acetyltransferase was purified to homogeneity using ammonium sulfate fractionation, followed by chromatography on DEAE-Sephadex (A-25), hydroxyapatite, Sephadex G-150, immunoabsorption and Sepharose-CoA columns. A purification of 3500-fold was achieved and the final preparation had a specific activity of 2:32 ?mol acetylcholine ...

Choline acetyltransferase immunoreactivity in the cat cerebellum
Neuroscience, 1991Choline acetyltransferase immunoreactivity was demonstrated in particular projection systems in cat cerebellum by combining immunohistochemistry, retrograde tracing and lesioning paradigms. The monoclonal antibody used in this study recognized a 68,000 mol. wt protein on immunoblots of cat cerebellum and striatum.

[247] Microdetermination of acetylcholinesterase, choline acetyltransferase, and choline
1971Publisher Summary This chapter discusses microdetermination of acetylcholinesterase, choline acetyltransferase, and choline. Labeled acetate produced by the enzymatic hydrolysis of acetylcholine is extracted into an organic solvent and counted.

Regulation of the Choline Acetyltransferase Gene
1992Genomic clones of human and porcine choline acetyltransferase were obtained using synthetic oligonucleotides to screen genomic libraries. Comparison of the human and porcine genes indicates a large degree of sequence conservation as well as a conservation of intron/exon structure. The human gene does not contain the ATG codon known to correspond to the
