Patient‐specific pharmacogenomics demonstrates xCT as predictive therapeutic target in colon cancer with possible implications in tumor connectivity

open access: yesMolecular Oncology, EarlyView.
This study integrates transcriptomic profiling of matched tumor and healthy tissues from 32 colorectal cancer patients with functional validation in patient‐derived organoids, revealing dysregulated metabolic programs driven by overexpressed xCT (SLC7A11) and SLC3A2, identifying an oncogenic cystine/glutamate transporter signature linked to ...

Management of E. coli sister chromatid cohesion in response to genotoxic stress

open access: yesNature Communications, 2017
Homologous recombination of DNA lesions in bacteria involves sister chromatid pairing. Here, the authors show that RecN promotes contacts between sister chromatids and facilitates repair.

Non-random distribution of high density chromatin detected at opposite ends of T-banded human metaphase chromosomes

open access: yesGenetics and Molecular Biology, 2007
Previous research using microdensitometric scanning and computer graphic image analysis showed that T-banded segments of human metaphase chromosomes usually exhibit an asymmetrical distribution of high density (HD) chromatin between sister chromatids ...

Targeted sister chromatid cohesion by Sir2. [PDF]

open access: yesPLoS Genetics, 2011
The protein complex known as cohesin binds pericentric regions and other sites of eukaryotic genomes to mediate cohesion of sister chromatids. In budding yeast Saccharomyces cerevisiae, cohesin also binds silent chromatin, a repressive chromatin ...

Survivin and Aurora Kinase A control cell fate decisions during mitosis

open access: yesMolecular Oncology, EarlyView.
Aurora A interacts with survivin during mitosis and regulates its centromeric role. Loss of Aurora A activity mislocalises survivin, the CPC and BubR1, leading to disruption of the spindle checkpoint and triggering premature mitotic exit, which we refer to as ‘mitotic slippage’.

Genetic attenuation of ALDH1A1 increases metastatic potential and aggressiveness in colorectal cancer

open access: yesMolecular Oncology, EarlyView.
Aldehyde dehydrogenase 1A1 (ALDH1A1) is a cancer stem cell marker in several malignancies. We established a novel epithelial cell line from rectal adenocarcinoma with unique overexpression of this enzyme. Genetic attenuation of ALDH1A1 led to increased invasive capacity and metastatic potential, the inhibition of proliferation activity, and ultimately ...

In Favor of Establishment: Regulation of Chromatid Cohesion in Plants

open access: yesFrontiers in Plant Science, 2017
In eukaryotic organisms, the correct regulation of sister chromatid cohesion, whereby sister chromatids are paired and held together, is essential for accurate segregation of the sister chromatids and homologous chromosomes into daughter cells during ...

Biomechanics‐Driven 3D Architecture Inference from Histology Using CellSqueeze3D

open access: yesAdvanced Science, EarlyView.
CellSqueeze3D reconstructs 3D cellular architecture from standard 2D histology images using biomechanical constraints and optimization. Validated on clinical datasets, it enables accurate tissue phenotyping, predicts gene mutations, and reveals significant correlations between nuclear‐cytoplasmic ratio entropy and tumor progression.

A requirement for STAG2 in replication fork progression creates a targetable synthetic lethality in cohesin-mutant cancers

open access: yesNature Communications, 2019
Cohesin complex mediates cohesion of sister chromatids and DNA loop formation. Here the authors show another role of cohesin in replication fork progression, suggesting a potential therapeutic strategy for cohesin-mutant cancers.
