The Replication of Yeast Chromosomes

1996
Eukaryotic chromosomes consist of linear DNA molecules that are complexed with proteins to form chromatin. Early analysis of replicating chromosomal DNA by fiber autoradiography revealed that replication initiates at multiple sites along each DNA molecule and that replication forks move bidirectionally away from the initiation sites (Huberman and Riggs

Bacterial chromosomes and their replication

This chapter reviews the current knowledge on the organization and replication of bacterial genomes with a special emphasis on the initiation of DNA replication. Most bacteria contain one chromosome, however a few species, like the human pathogen Vibrio cholerae, carry a secondary chromosome.

Origins of Chromosome Replication

1998
The first obligatory step for replication of duplex DNA is strand separation. Duplex melting (unwinding) leading to initiation of bacterial chromosome replication normally occurs at a fixed site on the chromosome termed oriC. This local duplex unwinding is effected by specific binding of an initiator, DnaA protein encoded by the dnaA gene, to 9-bp ...
