Structural and biochemical analysis of a B12 superbinder

BtuG proteins are vitamin B12 scavengers in Bacteroides thetaiotaomicron, a dominant human gut bacterium. We present crystal structures of three BtuG homologs bound to cobalamin and its precursor cobinamide, revealing picomolar binding affinities, among the highest known for any natural protein.

An Algebraic Circle Method

In this thesis we present an adaptation of the Hardy-Littlewood Circle Method to give estimates for the number of curves in a variety over a finite field. The key step in the classical Circle Method is to prove that some cancellation occurs in some exponential sums. Using a theorem of Katz, we reduce this to bounding the dimension of some singular loci.

L‐aspartate oxidase provides new insights into fumarate reduction in anaerobic darkness in Synechocystis sp. PCC6803

Synechocystis strains deficient in succinate dehydrogenase (SDH) secrete more succinate than the WT under dark anaerobic conditions, supporting that SDH then primarily acts as SDH, not as a fumarate reductase. L‐aspartate oxidase (Laspo) from Synechocystis is functional under anaerobic conditions, reducing fumarate to succinate.

Leucine‐rich glioma inactivated 1 (LGI1) is a ganglioside‐binding protein

A context‐dependent modulatory role for eIF6 in acquired resistance to vemurafenib in melanoma

Acquired resistance to vemurafenib upregulates the translation factor eIF6 in melanoma cells. Silencing eIF6 in resistant cells reduces proliferation and partially restores drug sensitivity, whereas its overexpression increases sensitivity across melanoma lines regardless of BRAF status, via modulation of mTOR, S6K, and MAPK signaling.

The role of miR‐335‐5p in the redifferentiation of BRAF p.V600E thyroid cancers

The BRAF p.V600E mutation promotes thyroid cancer dedifferentiation and radioiodine resistance. Using a network approach, we identified miR‐335‐5p as a key regulator of BRAF‐mutated thyroid tumors. Restoring miR‐335‐5p increased thyroid‐specific gene expression and iodine uptake in cells and organoids.
