Orthotopic Esophageal Cancer Xenograft Model in Immunosuppressed Microminipigs for Near‐Infrared Fluorescence Endoscopy

We report the first successful orthotopic engraftment of human esophageal squamous cell carcinoma in immunosuppressed microminipigs and demonstrate high‐contrast near‐infrared fluorescence endoscopic detection using EGFR‐targeted cetuximab. This large‐animal model bridges preclinical imaging research and future clinical translation in esophageal cancer.

A Zeolitic Pyrimidine Framework (ZPF) Nanoplatform Co‐Delivers a DNAzyme and a Protein Prodrug for Cascade‐Activated Tumour Therapy

A zeolitic framework co‐delivers a DNAzyme and a protein prodrug to tumour cells and executes a self‐amplifying therapeutic cascade: the DNAzyme elevates tumour oxidative stress to activate a caged RNase A prodrug. This spatiotemporally controlled synergy converts a genetic input into potent enzymatic cytotoxicity, suppressing tumour growth.

A noncanonical role for Jagged1 in endothelial mechanotransduction

The FEBS Journal, EarlyView.
This study reveals a noncanonical role for Jagged1 in endothelial mechanotransduction. Shear stress modulates Jagged1 expression and subcellular localization. Loss of Jagged1 attenuates mechanotransduction and reduces Src, VEGFR2, and ERK signaling. Direct mechanical stimulation of Jagged1 induces activation of these signaling pathways.

Quantitative analysis of DNA‐GATA1 binding alterations linked to hematopoietic disorders

Native holdup allows the quantitative determination of affinities between full‐length transcription factors and DNA. Mutations in either the protein or the DNA can modulate binding strength, which can be precisely quantified using this approach. Applied to GATA1, it revealed mutations that alter DNA binding.
