Effects of surrounding sequence on the suppression of nonsense codons
Journal of Molecular Biology, 1983Using a lacI-Z fusion system, we have determined the efficiency of suppression of nonsense codons in the I gene of Escherichia coli by assaying beta-galactosidase activity. We examined the efficiency of four amber suppressors acting on 42 different amber (UAG) codons at known positions in the I gene, and the efficiency of a UAG suppressor at 14 ...

How stop codon pseudouridylation induces nonsense suppression
Computational and Theoretical Chemistry, 2021Abstract Nonsense mutation can be an inherited disorder or caused by genetic mutation under certain conditions such as stress or exposure. Nonsense suppression, can have a broad potential as a therapeutic approach to nonsense mutation. Pseudouridine is proposed to result in nonsense suppression if induced in one of the three nonsense codons, Amber ...

TheBRCA2polymorphic stop codon: stuff or nonsense?
Journal of Medical Genetics, 2015Despite classification of the BRCA2c.9976A>T, p.(Lys3326Ter) variant as a polymorphism, it has been associated with increased risks of pancreatic, lung, oesophageal and breast cancer. We have noticed multiple co-occurrences of the BRCA2 c.9976A>T variant with the pathogenic BRCA2c.6275_6276delTT frameshift mutation p.(Leu2092ProfsTer7) and using a ...

Identification of nonsense codons in yeast
Journal of Molecular Biology, 1969Abstract Two nonsense codons have been found in Saccharomyces cerevisiae . The codons differ by a single base and only one contains guanine. Accepting the universality of the genetic code, one can deduce that these are amber and ochre codons since there exist suppressors capable of acting upon either class of nonsense alleles.

Base composition of nonsense codons in Escherichia coli
Journal of Molecular Biology, 1967Earlier oxperiments showed that the triplet UAG was the nonsense condon produced by the N1 class of alkaline phosphatase nonsense mutations. In the present experiments UAA has been identified as a second nonsense codon produced by the N2 class of alkaline phosphatase nonsense mutations.

The association of nonsense codons with exon skipping
Mutation Research/Reviews in Mutation Research, 1998Some genes that contain premature nonsense codons express alternatively-spliced mRNA that has skipped the exon containing the nonsense codon. This paradoxical association of translation signals (nonsense codons) and RNA splicing has inspired numerous explanations. The first is based on the fact that premature nonsense codons often reduce mRNA abundance.

Yeast mitochondrial tRNATrp can recognize the nonsense codon UGA
Nature, 1980DNA sequence analysis of mitochondrial genes that code for some mitochondrial proteins has suggested that the opal terminator, UGA, is used as a sense codon in mitochondria. The complete sequences of both the yeast and human genes coding for cytochrome oxidase subunit II contain UGA codons in the reading frame.

Reversion of trpA nonsense mutations by deletion of the chain-termination codons
Biochimie, 1989This paper describes a novel mechanism for reversion of nonsense mutations in the trpA gene of Escherichia coli. This mechanism, deletion of the nonsense codon, was discovered in the course of selecting for missense revertants of trpA(UGA211) and for catalytically active tryptophan synthetase alpha chain revertants of trpA(UAA234) and trpA(UAG234 ...

Orientation of Nonsense Codons on the Genetic Map of the Lac Operon
Science, 1967Intracodon recombination is used to orient the nonsense codons UGA, UAG, and UAA on the genetic map of the (lactose) lac operon of Escherichia coli . The 5′-end of these triplets is toward the operator end of the operon.
