Single‐molecule DNA flow‐stretch assays for high‐throughput DNA–protein interaction studies

open access: yesFEBS Open Bio, EarlyView.
We describe an optimised single‐molecule DNA flow‐stretch assay that visualises DNA–protein interactions in real time. Linear DNA fragments are tethered to a surface and stretched by buffer flow for fluorescence imaging. Using λ and φX174 DNA, this protocol enhances reproducibility and accessibility, providing a versatile approach for studying diverse ...

The Effect of pH and Color Stability of Anthocyanin on Food Colorant

open access: yesIOP Conference Series: Materials Science and Engineering, 2017
Anthocyanins are naturally occurring pigments of red and purple. Red anthocyanin pigments provide a strong and sharp and widely applied in various industries such as food coloring or drink. Anthocyanins isolated by maceration, extraction and thin layer chromatography (TLC).

Chameleon sequences reveal structural effects in proteins representing micelle‐like distribution of hydrophobicity

open access: yesFEBS Open Bio, EarlyView.
Amino acids sequence of two different proteins with the same sequence (chameleon sequence—black boxes) represent in 3D structure of the proteins different secondary structures: HHHH—helical and BBB—Beta‐structural. The chains folded in water environment adopt different III‐order structures in which the chameleon fragments appear to adopt similar status

Establishing an assay to evaluate d‐amino acid oxidase enzyme kinetics and inhibition using WST‐8 redox dye

open access: yesFEBS Open Bio, EarlyView.
This study investigated a novel WST‐8‐based assay for evaluating d‐Amino acid oxidase (DAO) inhibitors. We confirmed its effectiveness using known inhibitors and found that uremic toxins possess relatively weak inhibitory activity compared to existing drugs.

Analysing the significance of small conformational changes and low occupancy states in serial crystallographic data

open access: yesFEBS Open Bio, EarlyView.
This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

FEEDING SYSTEMS AND FRESH LAMB COLOR STABILITY

open access: yes
Consumers consider the surface color of fresh lamb as the indicator of freshness and wholesomeness, which affects their purchase decisions. The redox chemistry of myoglobin (Mb) determines the color of fresh lamb.

Screening and epitope characterization of Nidogen‐2‐specific nanobodies

open access: yesFEBS Open Bio, EarlyView.
Camel immunization and phage display were employed to generate high‐affinity VHH nanobodies against Nidogen‐2. After library construction, biopanning, ELISA screening, sequencing, and recombinant expression, selected nanobodies were purified and characterized, leading to the preliminary exploration of a nanobody‐based sandwich ELISA for specific ...

Color Changes of a Heat-Polymerized Polymethyl-Methacrylate (PMMA) and Two 3D-Printed Denture Base Resin Materials Exposed to Staining Solutions: An In Vitro Spectrophotometric Study

open access: yesApplied Sciences
Newly developed 3D-printed polymer materials are used for denture base fabrication. The aim of the present study was to evaluate the color stability of two new 3D-printed resins, a hard PPMA-based and a soft Urethane-based resin, in relation to a ...
