This protocol paper outlines methods to establish the success of a time‐resolved serial crystallographic experiment, by means of statistical analysis of timepoint data in reciprocal space and models in real space. We show how to amplify the signal from excited states to visualise structural changes in successful experiments.

Heuristic branch-and-price for building long term trainee schedules. [PDF]
Branch-and-price is an increasingly important technique for solving large integer programming models. Staff scheduling has been a particularly fruitful area since these problems typically exhibit a decomposable structure.

A Simple Perspective on Simultaneous Column and Row Generation [PDF]
To solve a linear programming problem with many variables and constraints, a simultaneous column and row generation algorithm can be used. Such an algorithm iteratively solves a restricted version of a linear programming problem including only a subset ...

Screening and epitope characterization of Nidogen‐2‐specific nanobodies
Camel immunization and phage display were employed to generate high‐affinity VHH nanobodies against Nidogen‐2. After library construction, biopanning, ELISA screening, sequencing, and recombinant expression, selected nanobodies were purified and characterized, leading to the preliminary exploration of a nanobody‐based sandwich ELISA for specific ...

A column generation approach to solve the crew re-scheduling problem
When tracks are out of service for maintenance during a certainperiod, trains cannot be operated on those tracks. This leads to amodified timetable, and results in infeasible rolling stock andcrew schedules. Therefore, these schedules need to be repaired.

Cytosolically synthesized chloroplast preproteins are translocated across the outer and inner envelope membranes through translocons called TOC and TIC, respectively. In green algae and plants, the TIC core is composed of essential membrane proteins, Tic12, Tic20, and Tic214.

Rapid screening of staphylokinase protein variants using an unpurified cell‐free expression system
An unpurified cell‐free protein synthesis (CFPS) platform enables rapid functional screening of staphylokinase variants. Direct plasminogen‐activation assays performed in microplate format provide real‐time activity readouts, allowing rapid identification and ranking of variants with improved or reduced fibrinolytic activity without protein ...

Evolutionarily divergent DUF4465 domains have a common vitamin B12‐binding function
We show that DUF4465 family proteins, widespread across bacteria from gut microbiomes, hydrothermal vents, and soil, share a common vitamin B12‐binding function. These augmented β‐jellyroll proteins bind vitamin B12 via extended loops. Our findings establish sequence‐diverse DUF4465 proteins as a widespread class of B12‐binding proteins, highlighting ...

Systemic dysregulation of apolipoproteins in amyotrophic lateral sclerosis serum
Amyotrophic lateral sclerosis (ALS) is a fatal disease that damages motor neurons. This study found that people with ALS show significant changes in blood fats and the proteins that carry them. Several apolipoproteins were higher, lipid balances were altered, and normal protein–lipid relationships were disrupted.
