Update on male hormonal contraceptive agents

Expert Opinion on Investigational Drugs, 2004
There remains a need for new acceptable and effective male contraceptives to increase the choice for couples throughout the world. There have been no recent advances in available male contraceptive methods although a number of promising approaches have been identified, of which the hormonal approach is currently undergoing clinical investigation.

Newer Agents for Hormonal Contraception in the Male

Trends in Endocrinology & Metabolism, 2000
Efforts to create a hormonal contraceptive for men use testosterone to suppress the production of pituitary gonadotropins and, hence, spermatogenesis. However, conventional testosterone must be administered by frequent injection, and when given alone, is not 100% effective.

Endogenous Antispermatogenic Agents: Prospects for Male Contraception

Annual Review of Pharmacology and Toxicology, 1978
A review of endogenous antispermatogenic agents as prospects for male contraception is reported. It is demonstrated that endogenous compounds exert regulatory influences at 4 major levels in the male: 1) between germ cells; 2) between Sertoli and germ cells; 3) between Leydig cells and seminiferous tubules; and 4) between the central nervous system

Effects of the male contraceptive agent gossypol on meiotic chromosomes of the male rat

Cytogenetic and Genome Research, 1985
Synaptonemal complexes (SCs) of rat spermatocytes were analyzed in silver-stained meiotic preparations 10–24 days after treatment with gossypol acetic acid, 30 mg/kg/day, for 70 days. Gossypol did not affect SC formation or function, as judged by the absence of pairing anomalies, SC fragmentation, or presynaptic arrest.

Investigations on the use of testosterone oenanthate as a male contraceptive agent

Contraception, 1974
The effect of exogenous testosterone on male gonadal function was in vestigated by applying a practical dose schedule of weekly injections of testosterone enanthate. 7 healthy young males received 22 intramuscular injections of 250 mg testosterone enanthate once a week for a period of 21 weeks. Mean sperm counts were reduced from a pretreatment value

Prazosin, an adrenergic blocking agent inadequate as male contraceptive pill

Contraception, 1988
The purpose of this study was to investigate the efficacy and the acceptability of Prazosin as a male contraceptive pill. Acceptable antifertility drugs for men are proving difficult to produce, and the possibility of using pharmacological agents to block selectively or to inhibit normal sperm transport through the male genital tract is an interesting ...

Structure-Activity Studies of 2,3,4,4a,5,9b-Hexahydroindeno[1,2-c]pyridines as Antispermatogenic Agents for Male Contraception

Journal of Medicinal Chemistry, 1995
Analogs of (4aRS,5SR,9bRS)-2-ethyl-2,3,4,4a,5,9b-hexahydro-7-meth yl-5-p- tolyl-1H-indeno[1,2-c]pyridine (Sandoz 20-438, 10a; R1 = ethyl, R2 = R3 = methyl, R4 = H) have been synthesized and tested in mice for their ability to reduce testes weight and disrupt spermatogenesis.

The effect of the male contraceptive agent gossypol acetic acid on mouse bone marrow cells in vivo: Micronuclei and mitotic index

Contraception, 1993
In this work the genotoxic effect of gossypol acetic acid (gossypol) was evaluated by determining the frequency of micronuclei and mitotic index in male mouse bone marrow cells in vivo. Bone marrow cells were collected at 24th hour after the single intraperitoneal (20, 40, and 80 micrograms/g) administration of gossypol.

Synthesis and assessment of fertility-regulating potential of 2-(2″-chloroacetamidobenzyl)-3-(3′-indolyl) quinoline in adult rats as a male contraceptive agent

Contraception, 2008
The purpose of this study was to investigate the fertility-regulating potential of the compound 2-(2''-chloroacetamidobenzyl)-3-(3'-indolyl) quinoline in male rats.Rats of proven fertility were treated with the compound by oral gavage for 1 to 8 consecutive weeks.

Is high dosage testosterone an effective male contraceptive agent?

Fertility and Sterility, 1988
In male contraceptive trials, approximately half of normal men become azoospermic on high dosages of testosterone enanthate (TE), whereas the other half of men become severely oligozoospermic. To determine whether sperm function is reduced in men with severe oligozoospermia induced by TE, we studied sperm function in six normal men whose sperm counts ...
