Dual Lineages of Langerhans Cells Cooperate to Restore the Immune Barrier after Skin Injury

After skin injury, the epidermal immune barrier is rebuilt by two sources of Langerhans cells. Resident Langerhans cells first move into the wound during re‐epithelialization, guided by CXCR2 signaling. Later, recruited monocytes become long‐lived Langerhans cells.

Heat stress promotes WNT5A secretion from fibroblasts via METTL3/YTHDC1‐dependent m6A methylation. Subsequently, WNT5A binds to FZD10 on melanocytes, activating β‐catenin‐driven melanogenesis. The Gela/decitabine/La2(CO3)3 colloidal (GDLC) hydrogel developed here could reverse the epigenetic silencing of the GSDME gene by the synergistic interaction between DAC and La3+, while concurrently activating caspase‐3, thus facilitating pyroptosis induction under the tumor microenvironment.
