Radiation‐Triggered Chemokine‐Secreting Probiotics for Boron Neutron Capture Therapy Based Radioimmunotherapy

open access: yesAdvanced Science, EarlyView.
Engineered Escherichia coli Nissle 1917 with a tannic acid‐mediated BPA coating enables tumor‐targeted boron delivery for BNCT. The low‐level incidental radiation generated during BNCT activate a RecA‐driven circuit to induce CXCL10, while radiation‐enhanced bacterial colonization triggers in situ vaccination and systemic antitumor immunity.

Ribonucleotides Amplify Mitochondrial DNA‐Driven cGAS–STING Activation via FdUMP‐ribonucleotide Hybrid Particles to Potentiate Chemoimmunotherapy

open access: yesAdvanced Science, EarlyView.
This work develops a metabolic sharing strategy that employs cytidine monophosphate (CMP) to competitively reprogram floxuridine (FUDR) pyrimidine metabolism in colorectal tumor cells. Elevated intracellular FdUMP induces nucleotide imbalance, drives mitochondrial DNA release and cGAS‐STING innate immune cascade activation, and potentiates robust ...

A simple large-scale method for separating closed circular form DNA by gel electrophoresis

open access: yes
The covalently closed form of circular duplex SV40 DNA was separated from the open and linear form of SV40 DNA by agarose gel electrophoresis in a large-scale gel system.

Reversible, Chemically Gated FRET via Ligand‐Activated Acceptors

open access: yesAdvanced Science, EarlyView.
Fluorogen binding turns the compact FAST tag into a tunable FRET acceptor, with ligand chemistry programming spectral overlap and excited‐state lifetimes. Fluorescence‐lifetime imaging reads out energy transfer through the shortened donor lifetime, while simply adding or washing out the dye switches FRET on and off, reversibly mapping the supramodular ...

A simple method for the preparation of the covalently closed circular form of plasmid DNA.

open access: yesBioTechniques, 1990
We describe a simple, rapid, inexpensive method for isolation of covalently closed circular plasmid DNA. The method involves the electrophoresis of crude DNA preparations in an agarose gel, electrotransfer onto a dialysis membrane and elution of the highly purified circular covalently closed plasmid DNA.
