Detection of circulating tumor DNA in colorectal cancer patients using a methylation‐specific droplet digital PCR multiplex

open access: yesMolecular Oncology, EarlyView.
We developed a cost‐effective methylation‐specific droplet digital PCR multiplex assay containing tissue‐conserved and tumor‐specific methylation markers. The assay can detect circulating tumor DNA with high accuracy in patients with localized and metastatic colorectal cancer.

The Single-Index/Cox Mixture Cure Model [PDF]

open access: yesBiometrics, 2018
AbstractIn survival analysis, it often happens that a certain fraction of the subjects under study never experience the event of interest, that is, they are considered “cured.” In the presence of covariates, a common model for this type of data is the mixture cure model, which assumes that the population consists of two subpopulations, namely the cured

ORACLE INEQUALITIES FOR THE LASSO IN THE COX MODEL. [PDF]

open access: yesAnnals of Statistics, 2013
We study the absolute penalized maximum partial likelihood estimator in sparse, high-dimensional Cox proportional hazards regression models where the number of time-dependent covariates can be larger than the sample size. We establish oracle inequalities

Stable variable selection for right censored data: comparison of methods [PDF]

open access: yes, 2011
The instability in the selection of models is a major concern with data sets containing a large number of covariates. This paper deals with variable selection methodology in the case of high-dimensional problems where the response variable can be right ...

Transcriptional network analysis of PTEN‐protein‐deficient prostate tumors reveals robust stromal reprogramming and signs of senescent paracrine communication

open access: yesMolecular Oncology, EarlyView.
Combining PTEN protein assessment and transcriptomic profiling of prostate tumors, we uncovered a network enriched in senescence and extracellular matrix (ECM) programs associated with PTEN loss and conserved in a mouse model. We show that PTEN‐deficient cells trigger paracrine remodeling of the surrounding stroma and this information could help ...

Expected Survival in the Cox Model

open access: yesScandinavian Journal of Statistics, 1997
The survival in a group of patients is calculated using a Kaplan–Meier curve and the hypothesis that the survival can be predicted by a specified Cox hazard is studied. This Cox hazard is obtained from a previous study of similar patients. A simple test of the hypothesis is constructed by comparing a suitable average of the individual predicted ...

Multiple imputation approach for interval-censored time to HIV RNA viral rebound within a mixed effects Cox model [PDF]

open access: yes, 2018
This is the peer reviewed version of the following article: “Alarcón-Soto, Y, Langohr K., Fehér, C., García, F., and Gómez, G. (2018) Multiple imputation approach for interval-censored time to HIV RNA viral rebound within a mixed effects Cox Model ...

PARP inhibition and pharmacological ascorbate demonstrate synergy in castration‐resistant prostate cancer

open access: yesMolecular Oncology, EarlyView.
Pharmacologic ascorbate (vitamin C) increases ROS, disrupts cellular metabolism, and induces DNA damage in CRPC cells. These effects sensitize tumors to PARP inhibition, producing synergistic growth suppression with olaparib in vitro and significantly delayed tumor progression in vivo. Pyruvate rescue confirms ROS‐dependent activity.

Stratified Cox Model with Interactions in Analysis of Recurrent Events

open access: yesActa Universitatis Lodziensis. Folia Oeconomica, 2018
The purpose of this paper is the assessment of relative intensity of exit from registered unemployment by means of the analysis of recurrent survival episodes and the comparison of these results with the results obtained for an individual episode.

Mixed Discrete and Continuous Cox Regression Model [PDF]

open access: yesLifetime Data Analysis, 2003
The Cox (1972) regression model is extended to include discrete and mixed continuous/discrete failure time data by retaining the multiplicative hazard rate form of the absolutely continuous model. Application of martingale arguments to the regression parameter estimating function show the Breslow(1974) estimator to be consistent and asymptotically ...
