AbstractDimeric associations of the D1-D2-CP47 and D1-D2-CP47-CP43 complexes of Photosystem II from spinach were isolated and purified with sucrose density gradient centrifugation and gel filtration chromatography and analyzed by electron microscopy and image analysis. Images of both preparations show characteristic details in protein density.

Water splitting by Photosystem II (PS II) in photosynthesis is catalyzed by a Mn4CaO5 oxygen-evolving complex (OEC). The reaction center D1 protein provides most of the ligands to the OEC but its complete assembly requires the association of the CP43 core antenna subunit which provides CP43:Glu341 as an essential ligand.
